摘要:以山鸡椒无菌苗叶片、茎段为外植体诱导愈伤组织,通过愈伤组织分化诱导不定芽途径,建立山鸡椒植株再生体系。结果表明,山鸡椒茎段是诱导愈伤组织最佳外植体,愈伤组织诱导和不定芽分化的最适培养基为MS+2.0 mg·L-1BA+0.1 mg·L-1IBA,分化培养基中添加适当浓度的维生素C可减轻愈伤组织褐化程度;诱导不定芽生根较理想的培养基为1/2MS+0.2 mg·L-1IBA+0.4 mg·L-1NAA。同时,通过组织学观察了愈伤组织的结构和不定芽的发生。
A system for plantlet regeneration of Litsea cubeba (Lour.) Pers. was established via adventitious buds induction from callus using leaves and stems as explants. The results showed that the stems were the optimal explants for callus induction. The best medium for calli induction and adventitious buds induction was MS+2.0 mg.L-1 BA+0.1 mg.L-1 IBA. In the meantime, the darkening degree of callus would be mitigated by adding appropriate concentration of vitamin C to the medium of adventitious buds induction. Moreover, the most favorable medium of root regeneration was 1/2MS+0.2 mg.Ll IBA+0.4 mg.Ll NAA. The structure of calli and the formation of adventitious buds were discovered bv histological analysis.
