摘要:利用人工控制授粉获得的麻竹F1群体,通过RAPD标记按拟测交作图策略构建了麻竹的连锁图谱。试验过程中从250个10bp的随机引物中筛选出16个,对双亲及44个子代进行扩增,共获得101条谱带,其中46条为符合1∶1分离的拟测交位点。利用多点连锁分析,共形成7个连锁群,覆盖的总图距为305.7cM,最长为100.0cM,最短为12.4cM,标记间的平均图据约为12.74cM,从而为构建高密度的麻竹遗传图谱提供了基础。
F1 generation of Dendrocalamus latiflorus Mtmro was obtained from artificial pollination, and random amplified polymorphic DNAs (RAPDs) were used to construct linkage maps of the parents of them. 16 random primers were screened out from 250 ones to generate a sample of 44 F1 progenies and their parents, total 101 loci were identified, among which 46 were segregating 1:1 (testcross configuration).7 linkage groups were formed by using multiple analysis, the resulting linkage map of Dendrocalamus latiflorus Munro sparmed 305.7cM, with the largest of 100.0 cM and the shortest of 12.4 cM,the average distance is 12.74 cM, and this provide bases for construction a advanced genetic maps.
