摘要:为了揭示水杨酸甲酯转移酶SAMT (Salicylic acid carboxyl methyltransferase)基因在杨树抗溃疡病中的功能。本研究采用RT-PCR技术和pGM-T克隆的方法,获得了‘84K杨’的SAMT基因序列,并对该基因编码的蛋白的理化性质、疏水性、结构域、功能、二级结构及亚细胞定位等进行了生物信息学分析。结果显示:该基因位于细胞质中,属于甲基转移酶7家族,为亲水性蛋白。预测结果为下一步研究该基因在杨树抗溃疡病中的功能提供了帮助,也为其在其他多年生木本植物中的功能研究提供了参考。
In order to reveal the function of methyl salicylate transferase SAMT(Salicylic acid carboxyl methyltransferrase) gene in poplar anti-ulcerative disease.The SAMT gene sequence of ’Poplar 84 K’ was obtained by RT-PCR and p GM-T cloning.Meanwhile,the physicochemical properties,hydrophobicity,domain,function,secondary structure and subcellular localization of the protein encoded by the gene were obtained by bioinformatics analysis.The results showed that the gene was located in the cytoplasm and belonged to the methyltransferase 7 family,which was a hydrophilic protein.The prediction results provide a basis for the further study of the function of the SAMT gene in poplar canker resistant and also provide a reference for its functional studies in other perennial woody plants.
