摘要:探讨了用范氏法测定植物中酸性洗涤木质素(ADL)的影响因素,对不同消煮时间、不同硫酸浓度、不同浸泡时间条件下的测定结果进行了比较分析。结果表明:样品消煮的适宜时间为30 min,硫酸适宜浓度为72%~75%,浸泡适宜时间为3~4 h;人工检测结果的变异系数(CV)在2.23%~6.66%之间,仪器法的CV在0.15%~1.10%之间;人工检测与仪器法所测结果无显著性差异;范氏法与王玉万法所测结果无显著性差异,这两种方法适用于植物ADL的测定。
The author explored the factors affecting the determination of plant acid detergent lignin(ADL) by using Van Soest method, and compared and analyzed the determination results under different conditions. The results showed that: the suitable time for sample boiling was 30 min;the suitable concentration of sulfuric acid for sample decomposition was 72%~75%;the appropriate time for sample soaking was 3~4 h;the coefficient of variation(CV) of manual determination results was 2.23%~6.66%, while the CV of instrument determination results was 0.15%~1.10%;there was no significant difference in determination results between manual method and instrument method;there was also no significant difference in determination results between Van Soest method and Wang Yuwan’s method, and these two methods were all suitable for the determination of plant ADL.
