摘要:本研究采用Illumina高通量测序技术对3种不同倍性的桤木属植物混合样品进行转录组测序,利用生物信息学方法开展基因表达谱的研究、功能基因的预测。测序结果组装获得85 769个Unigene,总长50 200 910 bp,功能注释到NR、NT、Swiss-Prot、KEGG、COG、GO库的Unigene分别是45 172、38 673、29 181、25 096、14 968和30 675个,所有注释上的Unigene是48 204个。根据KEGG pathway数据库,对桤木转录组的Contig进行pathway生物学通路的注释和预测,共识别出25 096个Unigene具有对应的功能,并关联到128条生物学通路。SSR查找发现,从85 769个Unigene中找到8 678个SSR位点,占Unigene总数的比例为10.11%。其中,二核苷酸重复所占比例最高,达到65.87%,其次是三核苷酸重复,为28.36%,四、五、六核苷酸重复类型的数量很少,总计5.77%。SSR不同重复单元类型中,出现频率最高的为AG/CT,其次是AAG/CTT和AT/AT。
The transcriptome of the leaf of Alnus plants with different ploidy was sequenced by Illumina, a new generation of high-throughput sequencing technology was used to study the expression profiling and predict the functional gene. In the results of assembly, 85 769 Unigenes were detected, and a total length of 50 200 910 bp,and the total annotation Unigenes were 48 204. For function annotation analysis, we got 45 172, 38 673, 29 181,25 096, 14 968, 30 675 Unigenes which annotated to the NR, NT, Swiss-Prot, KEGG, GO database, respectively.A total of 25096 Unigenes were associated with 128 biological pathway by annotating and forecasting the biological pathways for the transcriptome of Alnus. 23 262 SSR in 85 769 Unigenes were found, which accounted for 27.12% proportion of the total number of Unigenes. The characteristic of SSR distribution showed that di-nucleotide repeat was the highest reaching 65.87%, following by tri-nucleotide repeat, which was 28.36%, the total proportion of tetra-nucleotide, penta-nucleotide and hexa-nucleotide only was 5.77%. The types of SSR were analyzed that AG/CT was the highest repeat, following by AAG/CTT and AT/AT.
