摘要:日本落叶松是落叶松属下的一个物种,是我国引种栽培的造林优良树种之一,不仅对中国的森林木材的供应,城市绿化等方面有重大影响,而且具有较高的生态价值。【目的】近年来松材线虫在我国逐渐向北方扩张的同时,在南方地区不断危害新的寄主。由于日本落叶松在我国湖北地区有较广的栽植面积,并且是松材线虫病的感病寄主之一,目前材线虫病疫情已扩散我国16个省(区、市)和316个县级行政区。至今松材线虫病发生已造成面积高达已超过8万hm^2的松木的死亡,对我国森林森态系统造成极大的威胁,由于日本落叶松在我国湖北地区有较广的栽植面积,并且是松材线虫病的感病寄主之一,因此对于湖北地区日本落叶松材线虫病的研究迫在眉睫。【方法】本研究在湖北省恩施州建始县长岭岗地区采集枯死的日本落叶松样本,利用显微镜形态观察和分子测序的方法对病原线虫进行综合鉴定。【结果】结果发现建始县长岭岗地区枯死的日本落叶松体内存在松材线虫,形态鉴定结果符合松材线虫形态特征,分子鉴定结果与NCBI数据库中松材线虫的ITS和28S多个序列基本相同。【结论】综合判断枯死日本落叶松中的病原线虫为松材线虫,由此确定松材线虫在我国湖北省恩施州建始县长岭岗地区自然条件下可侵染日本落叶松。为湖北地区日本落叶松的病虫害防控提供了新的参考。
Larix Kaempferi is one of the species under larch and one of the excellent afforestation trees introduced and cultivated in China.It not only has great influence on China’s forest timber supply,urban greening and other aspects,but also has high ecological value.【Objective】In recent years,pine wood nematode has gradually expanded to north in China and harmed new hosts in south.L.Kaempferi has been widely planted in Hubei province of China,and is one of the main hosts of pine wood nematode disease.So far,the occurrence of pine nematode disease has caused the death of more than 80,000 hm^2 of pine.It poses a great threat to the forest ecosystem in China.L.Kaempferi has been widely planted in Hubei province of china,and is one of the main hosts of pine wood nematode disease,so it is urgent to study the disease of Japanese larch nematode in Hubei area.【Method】The occurrence of pine wilt disease in the samples of L.kaempferi in Hubei province was detected.Nematodes isolated from dead L.kaempferi were also identified by microscopic observation and molecular sequencing.【Result】The results showed that the nematode found in the L.kaempferi has the same morphological characteristics as the pine wood nematode.The result of molecular identification indicated that the ITS and 28S sequences of pine wood nematode identified were basically same as the sequences in the NCBI database.【Conclusion】Through comprehensive judgment,the pathogenic nematode of dead L.kaempferi was pine wood nematode.The pine wood nematode began to invade L.kaempferi under natural conditions in Hubei.It provides a new reference for the prevention and control of diseases and insect pests of L.kaempferi under in Hubei.
