摘要:为了筛选获得高效拮抗建兰炭疽病病原菌胶孢炭疽菌(Colletotrichum gloeosporioides)的菌株,采用平板对峙法,从新疆库姆塔格沙漠土样分离的126株细菌中初筛获得23株拮抗菌株,经平板扩散法复筛得到1株高效拮抗菌株N147-1-3。结合菌落菌体形态观察、16S r DNA序列分析和生理生化试验,鉴定该菌为甲基营养型芽孢杆菌(Bacillus methylotrophicus)。对拮抗菌发酵液上清测定表明,60%的硫酸铵沉淀物具有最高的抑菌活性,经氯仿抽提后萃取相不具有拮抗活性,经高温处理和蛋白酶K消化后抑菌物质的拮抗作用消失,初步分析抑菌物质是蛋白类物质。
Twenty-three strains were obtained by screening strains with high antagonism against CoUetotri- chum gloeosporioides by the tablet confrontation method from 126 strains,which were isolated from Xin- jiang Kumtag desert samples. Strain N147-1-3 with highest antagonism was obtained by using the plate diffusion method. Combined with the morphology observation of colonies and cells, 16S rDNA sequence analysis ,physiological and biochemical characteristics tests, the strain was finally identified as Bacillus methylotrophicus. The test results of the supernatant of the strain fermented liquid showed that the sedi- ment by adding 60% ammonium sulfate had the highest antibacterial activity. The supernatant of the strain fermented liquid lost its antimicrobial activity after the chloroform extraction. The antagonism effect of the strain disappeared after treatments by high temperatures or proteinase K. The antibacterial sub- stance was finally identified as protein.
