摘要:为了实现防腐处理材中戊唑醇和丙环唑含量的准确快捷分析,本研究优化了防腐木中戊唑醇和丙环唑提取条件,建立戊唑醇和丙环唑的高效液相色谱(HPLC)分析方法。HPLC分析采用紫外检测器及C18反相色谱柱,以V_(甲醇)∶V_(乙腈)∶V_(水)=1∶1∶1为流动相,流速1.2 mL/min,检测波长220 nm。利用超声波提取法进行木材中防腐剂提取的预处理,并对提取条件进行优化。结果表明:在5~100 mg/L范围内,戊唑醇和丙环唑标准曲线的决定系数R 2均为0.9997,具有良好的线性相关性;分析样品的变异系数分别为0.98%和0.83%,样品的回收率为99.0%~101.3%和98.9%~100.8%。超声波提取法中,以甲醇或甲醇和水为溶剂提取效果均较好,选择甲醇和水体积比8∶2最为合适,可最大程度减少甲醇用量;溶剂用量和提取时间对结果均有一定影响,采用液料比30∶1(mL∶g)、提取时间30 min为最优条件。最优条件下,处理和分析了辐射松、杉木、橡胶木、杨木和竹材5种样品,所有木竹中戊唑醇和丙环唑的回收率分别为95.2%~102.1%和95.9%~104.0%,标准偏差分别为0.7%~3.0%和0.3%~2.8%。超声波提取预处理和HPLC分析方法可用于检测处理材中戊唑醇和丙环唑的含量,前处理程序简便快捷,分析结果准确度较高、重现性好。
Treatment of wood with preservatives can prolong the service life of wood products significantly.It is an important measure to save wood and possess great significance to the efficient use of wood resources.Copper azole(CA)is a water-borne wood preservative constituted by copper amine complex and triazole fungicide,wherein the triazole fungicide is tebuconazole(TEB)and propiconazole(PPZ).Furthermore,as ergosterol biosynthesis inhibitors,TEB and PPZ have a good control function on wood decay fungi such as Basidiomycetes and Ascomycetes,which have advantages of low cost and good environmental compatibility.The method to quality control of the treated wood is to analyze the content of TEB and PPZ accurately.TEB and PPZ are generally analyzed by the high performance liquid chromatography(HPLC),and fully extracting them from the treated wood is the key procedure to accurate analysis of their content.In this study,the extraction conditions of the tebuconazole and propiconazole from the treated wood were optimized,and an analytical method of TEB and PPZ by the HPLC was established.The UV detector and C18 reversed-phase chromatography column were used for detection.The chromatographic conditions for the analysis of the tebuconazole and propiconazole were as follows:the flow phase methanol-acetonitrile-water(1∶1∶1,v∶v∶v),flow rate 1.2 mL/min,and UV detection wavelength 220 nm.Meanwhile,the optimization of the ultrasonic extraction method on extracting TEB and PPZ from the radiata pine was conducted by the effects of different solvents,the amount of solvent and the extraction time.In the range of 5-100 mg/L,the correlation coefficients of tebuconazole and propiconazole were both 0.9997,with good linearity.The coefficients of variation of the analyzed sample were 0.98%and 0.83%,and the sample recovery rates were 99.0%-101.3%and 98.9%-100.8%,respectively.In the ultrasonic extraction method,the methanol or methanol-water as the solvent has better extraction effect,and the methanol-water ratio of 8∶2(v∶v)is the most suitable ratio,which can minimize the amount of methanol.The amount of solvent and extraction time have a certain influence on the results,and the optimal conditions were 30∶1(mL∶g)liquid-solid ratio and 30 min extraction time.Under optimal conditions,five samples of radiata pine,Chinese fir,rubber wood,poplar and bamboo were processed and analyzed.The recovery rates of the tebuconazole and propiconazole were 95.2%-102.1%and 95.9%-104.0%,with the standard deviations were 0.7%-3.0%and 0.3%-2.8%,respectively.This method can be used for analyzing the content of the tebuconazole and propiconazole in the treated wood materials,not only the pre-treatment procedure is simple and quick,but also the results are high accuracy and good reproducibility.
