摘要:以黑木相思(Acacia melanoxylon)优良单株(AMY12004)的当年新生枝条带腋芽茎段为外植体,灭菌后接入MS培养基上培养,以其无菌萌芽的叶片、茎段和叶柄为实验材料,通过间接器官发生途径建立黑木相思愈伤组织诱导及高频植株再生体系。研究结果表明,诱导愈伤组织的最佳外植体为茎段;愈伤组织诱导的最佳培养基为MS+1.5 mg·L–16-BA+0.2mg·L–1NAA+3%蔗糖,诱导率为93.33%;愈伤组织再分化的最佳培养基为MS+2.0 mg·L–16-BA+0.5 mg·L–1NAA+3%蔗糖,分化率为79.17%,再生系数为9.58;再生芽生根的最佳培养基为MS+0.5 mg·L–1IBA+0.5 mg·L–1NAA+4%蔗糖,生根率为96.05%,移栽存活率为81.40%。
Newly born stems with axillary buds of eminent Acacia melanoxylon(AMY12004) were used as explants and cultured on MS after sterilization. The aseptic leaf, petiole and stem of germinated axillaries were used to induce callus, and high-frequency regeneration systems were established via indirect organogenesis. The optimal material for callus induction was the stem, the optimal medium for induction was MS+1.5 mg·L–16-BA+0.2 mg·L–1NAA+3%sucrose, and the optimal medium for redifferentiation was MS+2.0 mg·L–16-BA+0.5 mg·L–1NAA+3%sucrose. The callus induction rate, redifferentiation rate and regeneration coefficient was 93.33%, 79.17% and 9.58, respectively. The optimal rooting medium was MS+0.5 mg·L–1IBA+0.5 mg·L–1NAA+4%sucrose and the highest rooting rate was 96.05%; the transplant survival rate was 81.40%.
