摘要:[目的]对平沙绿僵菌WP08的几丁质酶基因家族成员进行鉴定,解析其在不同培养条件下的表达模式,以期为深入研究平沙绿僵菌WP08几丁质酶基因的功能奠定基础。[方法]通过生物信息学对平沙绿僵菌WP08几丁质酶基因家族成员进行系统分析,利用实时荧光定量技术检测该家族成员在PPDA培养基和添加虫体粉末的诱导培养基MC中的表达模式。[结果]从平沙绿僵菌WP08基因组中鉴定出15个几丁质酶基因家族成员,编码181~1 322 aa,几乎所有成员都偏酸性,属于稳定蛋白,分布在8条染色体上,且均有内含子结构。种间共线性分析展示出平沙绿僵菌与白僵菌、蛹虫草的几丁质酶基因可能由相同祖先进化而来,系统进化分析说明平沙绿僵菌WP08几丁质酶基因家族主要包括3个亚组,其中亚组B的成员数量最多。平沙绿僵菌WP08不同亚组的几丁质酶成员在不同情况下的表达模式不同,亚组B几丁质酶基因在诱导表达培养基中呈现出高水平的表达;部分亚组A几丁质酶基因表达水平则在诱导培养基中下降。[结论]平沙绿僵菌WP08几丁质酶基因家族成员不同亚家族成员具有功能分化,其中亚组B几丁质酶成员在平沙绿僵菌侵染昆虫的过程中可能发挥了重要作用,该研究结果为后续以几丁质酶基因为靶点开发新的绿僵菌生防菌剂提供理论依据。
[Objective]To identify the chitinase gene family members in Metarhizium pingshaense strain WP08 and analyze the expression patterns of these chitinase genes in different substrate,for providing a foundation for further study on the function of chitinase genes in M.pingshaense strain WP08.[Methods]The chitinase family members of M.Pingshaense strain WP08 were systematically analyzed by bioinform-atic methods.Real-time fluorescence quantitative technique were used to detect the expression patterns of chitinase family members in PPDA medium and MC induction medium which were added with insect powder.[Results]A total of 15 members of chitinase gene family were recognized from the genome of M.pingshaense strain WP08,encoding proteins from 181 to 1322 amino acids.Most of these proteins were acidic and stable,distributed across 8 chromosomes,and contained introns.Interspecific collinearity ana-lysis showed that the chitinase gene of M.pingshaense,Beauveria bassiana and Cordyceps militaris may have evolved from the same ancestor.Phylogenetic analysis indicated that the chitinase gene family of M.pingshaense strain WP08 could be divided into three subgroups,with subgroup B constraining the largest number of members.Expression analysis revealed distinct expression patterns among different subgroups under various conditions.Chitinase genes in subgroup B showed high level of expressions in the induced medium,whereas the expression of certain subgroup A chitinase genes declined under the same condi-tions.[Conclusion]Different subfamily members of the chitinase gene family in M.pingshaense strain WP08 have functional differentiation,and the subgroup B chitinase genes of Metarhizium Pengshaense may play an important role in the infection with insects.The findings provide a theoretical basis for the de-velopment of novel biological inoculants targeting chitinase genes in M.pingshaense.
