摘要:为挖掘杜仲嫩枝扦插生根过程中关键long non-coding RNAs(lncRNAs),本研究以‘华仲6号’和‘华仲8号’杜仲嫩枝扦插后0、18和32 d插穗基部的皮层组织为材料,构建链特异性文库并进行测序分析。结果显示,12个样品共获得5862个lncRNAs和437个差异表达lncRNAs,且差异表达lncRNAs分别通过顺式和反式调控1050和5002个靶基因,功能富集分析显示这些靶基因主要参与植物激素信号转导、MAPK信号通路、淀粉和蔗糖代谢等通路。在植物激素信号转导通路中,鉴定了ARF、AUX/IAA、SAUR、PP2C、JAR、GID1和TGA等基因。最后构建了差异表达lncRNAs与靶基因调控网络,发现MSTRG.33055.1、MSTRG.36256.1、MSTRG.36256.2、MSTRG.51731.1和MSTRG.33079.1通过调控生长素和脱落酸相关基因的表达,进而影响不定根的形成。研究结果为深入研究杜仲插穗不定根形成的分子机制提供参考,同时完善了杜仲无性繁殖的理论基础。
To identify the key long non-coding RNAs(lncRNAs)involved in adventitious root formation of Eucommia ulmoides softwood cuttings,this study constructed strand-specific libraries and conducted sequencing analysis using the cortical tissues from the base of cuttings of‘Huazhong 6’and‘Huazhong 8’E.ulmoides at 0,18 and 32 days after cutting.The results showed that a total of 5862 lncRNAs and 437 differentially expressed lncRNAs were obtained from 12 samples.These differentially expressed lncRNAs regulated 1050 and 5002 target genes through cis-and trans-action,respectively.Functional enrichment analysis showed that these target genes were mainly involved in plant hormone signaling transduction,MAPK signaling pathway,starch and sucrose metabolism pathway,etc.In the plant hormone signaling pathway,ARF,AUX/IAA,SAUR,PP2C,JAR,GID1,and TGA were identified.Finally,the differentially expressed lncRNAs and target genes regulatory networks were constructed.MSTRG.33055.1,MSTRG.36256.1,MSTRG.36256.2,and MSTRG.33079.1 regulated the expression of auxin and abscisic acid related genes,thereby affecting the formation of adventitious roots.The results will provide a reference for further research on the molecular mechanism of adventitious root formation of E.ulmoides cuttings,and also improve the theoretical basis of the vegetative propagation of E.ulmoides.
