摘要:研究了 2株相思根瘤菌在 4种培养基中的生长情况 ,并在此基础上应用正交实验L9(34 )对相思根瘤菌进行优化培养基研究 ,筛选出相思根瘤菌优化培养基配方。结果表明 :MZ菌株与YM2菌株在各自优化培养基的菌体数量高达 8.2× 10 9个 mL与 6 .3× 10 9个 mL ,分别为原有培养基的 3.6 4倍与 1.14倍 ;另外用乙炔还原法测得它们的最高固氮酶活性分别为 0 .96 3μg .mL- 1 .d- 1 与 0 .2 80 μg .mL- 1 .d- 1 。
The growth behavior of Acacia Rhizobium(MZ and YM2) in four mediums were studied in this paper.Accordingly,their optimized mediums were obtained from L_9(34)orthogonal test.The results showed that the number of MZ and YM2 were respectively 8.2×109/mL and 6.3×109/mL in their own optimized medium,which were 3.64 and 1.14 times of their conventional mediums.The study of nitrogenase activity by Acetylene reduction method showed that MZ and YM2 had comparatively high nitrogen-fixation activity,which reached 0.963μg.mL(-1).d(-1) and 0.280 μg.mL(-1).d(-1).
