摘要:查尔酮合成酶(chalcone synthase,CHS)是Ⅲ型聚酮合酶的关键成员,该酶催化类黄酮生物合成途径的第一步,是苯丙氨酸代谢途径过程中的重要限速酶。为了进一步探索CHS基因家族在沉香形成中的作用,本研究对11个白木香CHS(AsCHS)基因编码蛋白的理化性质、亚细胞定位、蛋白结构、系统进化以及基因组织表达和伤害处理表达等方面进行了系统研究。结果显示:11个AsCHS基因均含有CHS-Like保守序列;大部分As CHS蛋白为亲水性蛋白;As CHS1~AsCHS4为稳定蛋白,其余不稳定;As CHS蛋白的二级结构主要由无规卷曲与α-螺旋结构组成;As CHS蛋白均不含信号肽;亚细胞定位分析表明大部分As CHS蛋白位于细胞质。在系统进化分析中,白木香CHS基因家族中11个成员被划分为3组。CHS基因家族成员在白木香不同组织中具有不同的表达模式;AsCHS基因对钻孔处理均有所响应。本研究对白木香CHS基因家族进行鉴定和生物信息学分析,为深入研究白木香CHS基因家族功能提供了一定的参考和依据。
Chalcone synthase(CHS)is a key member of typeⅢpolyketide synthase,which catalyzes the first step of flavonoid biosynthesis and is an important rate-limiting enzyme in the process of phenylalanine metabolism.In order to further explore the role of the CHS gene family in the formation of agarwood,this study investigated the physicochemical properties,subcellular localization,protein structure,phylogenetic evolution,gene organization expression,and injury treatment expression of the proteins encoded by 11 CHS genes from Aquilaria sinensis(AsCHS).The results showed that:all the 11 AsCHS genes contained CHS-Like conserved sequences;most AsCHS proteins were acidic and hydrophilic proteins;AsCHS1~AsCHS4 were stable proteins,and the rest were unstable;the secondary structure of AsCHS proteins were mainly composed of random coil andα-helix;AsCHS proteins do not contain signal peptides;subcellular localization analysis showed that most AsCHS proteins are located in the cytoplasm.In the phylogenetic analysis,the CHS gene family were divided into 3 groups.CHS gene family members of A.sinensis had different expression patterns in different tissues;AsCHS all responded to drilling treatment.In this study,the identification and bioinformatics analysis of the CHS gene family provided a certain reference and basis for further research on the function of the CHS gene family of A.sinensis.
