摘要:以观赏型良种‘华仲11号’杜仲(Eucommia ulmoides‘Huazhong No.11’)及其叶片呈皱缩性状的芽变杜仲为研究对象,提取DNA进行10倍的全基因组重测序。对比‘华仲11号’杜仲及其芽变杜仲在叶片形态形成、激素合成方面的变异位点,分析相关变异位点的生物学意义,筛选叶片皱缩形状相关的基因,探索与杜仲(Eucommia ulmoides)叶片皱缩性状紧密联系的单核苷酸多态性(SNP)位点,筛选杜仲叶片性状相关变异基因。结果表明:‘华仲11号’杜仲及其芽变杜仲,与参考基因组之间均发生单核苷酸多态性、小片段的插入和缺失(InDel)、染色体结构变异(SV)。在杜仲参考基因组(1.18×10^(9))基础上,芽变杜仲具有5732071个单核苷酸多态性、293566个小片段的插入和缺失、3722个染色体结构变异;‘华仲11号’具有5711141个单核苷酸多态性、293078个小片段的插入和缺失、3870个染色体结构变异。通过‘华仲11号’杜仲及其芽变杜仲之间的比较发现,含有变异位点的26722条基因与蛋白功能变异有关;根据数据库功能注释结果,共筛选出13条有关基因及30个单核苷酸多态性位点。
In order to discover the SNP sites closely related to the leaf shrinkage traits of Eucommia ulmoides(E.ulmoides‘Huazhong No.11’)and screen the variant genes related to the leaf traits of E.ulmoides,we took the ornamental variety‘Huazhong 11’and its bud change E.ulmoides with shriveled leaves as the object,extracted DNA and resequencing the whole genome of 10×.The mutation sites in leaf morphogenesis and hormone synthesis of‘Huazhong 11’and its bud change E.ulmoides were compared,the biological significance of the mutation sites was analyzed,and the genes related to leaf shrinkage shape were screened.The single nucleotide polymorphisms(SNP),small insertions and deletions(InDel)and chromosomal structural variation(SV)occurred between the reference genome and both of‘Huazhong 11’and its bud change E.ulmoides.Fron the reference E.ulmoides genome(1.18×10^(9)),the budding E.ulmoides has 5732071 SNPs,293566 InDels,and 3722 SVs;‘Huazhong 11’has 5711141 SNPs,293078 InDels,and 3870 SVs.Through the comparison between‘Huazhong 11’and its bud change Eucommia ulmoides,it was found that 26722 genes with mutation sites are related to protein function variation.From the results of the database function annotation,a total of 13 related genes and 30 SNPs were screened out.
