摘要：为探究柿叶红茶和柿叶绿茶抗氧化活性差异及代谢物特征,采用紫外-可见分光光度法测定其总酚、总黄酮含量及抗氧化活性,并结合超高液相-串联质谱(UPLC-MS)对代谢物进行筛选和鉴定。结果表明:柿叶红茶总酚、总黄酮含量及DPPH自由基、ABTS自由基清除能力均显著高于柿叶绿茶;UPLC-MS共鉴定出199个差异代谢物,以类黄酮、生物碱、有机酸及其衍生物、核苷酸及其衍生物、氨基酸及其衍生物、脂质、酚酸7类为主,占比分别为22.61%、9.55%、8.04%、8.04%、7.04%、7.04%、6.53%,其中类黄酮、生物碱类和酚酸类物质的组成和含量差异,可能是构成柿叶红茶和柿叶绿茶差异的重要原因。
To investigate the differences in antioxidant activities and metabolite characteristics between persimmon leaf black tea and green tea,the total polyphenol,flavonoid contents,and antioxidant activities were determined by UV-visible spectrophotometry,and the metabolites were screened and identified in combination with ultra-high liquid chromatography-tandem mass spectrometry(UPLC-MS).The results showed that the total polyphenol,flavonoid contents,and the antioxidant activities of DPPH and ABTS radicals of persimmon leaf black tea were significantly higher than those of persimmon leaf green tea;a total of 199 differential metabolites were identified by UPLC-MS,with 7 categories,namely flavonoids,alkaloids,organic acids and their derivatives,nucleotides and their derivatives,and amino acids and their derivatives,lipids and phenolic acid,accounting for 22.61%,9.55%,8.04%,8.04%,7.04%,7.04%and 6.53%,respectively,of which the differences in the composition and content of flavonoids,alkaloids and phenolic acids may be an important reason for the differences of persimmon leaf black tea and persimmon leaf green tea.
