摘要:建立杨树高效的植株再生体系是采用生物技术手段对其进行遗传改良的基础和前提。该研究以山新杨无菌苗叶片为外植体材料,通过对附加不同浓度BA和NAA的MS培养基的筛选,建立了不定芽的高频再生体系,其再生率、生根率及移栽成活率均达到100%。在此基础上,确定了不定芽再生和生根的卡那霉素选择压。对再生苗的RAPD分析结果表明,该再生体系基本能够保持原有供体植株的遗传基础,可以应用于试管苗快速无性繁殖及遗传转化。
The establishment of a highly efficient plant regeneration system is a basic precondition for genetic improvement of poplar by biotechnology.A highly efficient plant regeneration system with 100% shoot regeneration,rooting and survival rate from leaf explants of Populus davidiana×P.bolleana in vitro was established by comparing different concentrations of BA and NAA on MS medium.In the experiments,the optimal selection pressures of kanamycin for shoot regeneration and rooting were also determined.The analysis of RAPD on 15 random selected regenerated plants revealed that over ninety percent of regenerated plants kept the genetic basis of the donor plants.Results of this study can be applied to micropropagation and genetic transformation of P.davidiana×P.bolleana.
