摘要:[目的]明确2株昆虫病原真菌的种类及生物学特性,测定它们对松墨天牛(Monochamus alternatus Hope)幼虫的侵染活力。[方法]对2株不同寄主和地区来源的昆虫病原真菌进行分离和纯化,并基于形态学特征及r DNA-ITS、TEF-1α和Bloc序列的同源性明确其分类地位。观察并测定2菌株的分生孢子大小、菌落生长速率、产孢量、孢子萌发率以及对松墨天牛3龄幼虫的生物活性。[结果]形态学观察及多基因序列比对分析结果显示,2株昆虫病原真菌均为球孢白僵菌(Beauveria bassiana),分别编号为LCL-8和MLY-9。其分生孢子均为圆球形或近圆球形,大小分别为(2.414±0.210)μm和(2.681±0.225)μm。生物学特性测定结果表明,LCL-8和MLY-9在PDA培养基(25℃)上的菌落生长速率分别为(4.050±0.052) mm·d^(-1)和(3.825±0.022) mm·d^(-1),产孢量分别为(8.504±0.814)×10^(7)孢子·cm^(-2)和(7.592±0.260)×10^(7)孢子·cm^(-2),24 h孢子萌发率分别为(82.2±4.3)%和(93.2±3.2)%。其中,2菌株在孢子大小、菌落生长速率和24 h孢子萌发率上存在显著性差异(p<0.05)。生测结果显示,LCL-8和MLY-9对松墨天牛3龄幼虫均具有强致病力,且MLY-9的毒力更高。当浓度为1×10^(8)孢子·mL^(-1)时,LCL-8和MLY-9对试虫的累计校正死亡率分别在第11 d和第8 d达到100.0%,其LT50分别为5.470 d和5.335 d。此外,LCL-8和MLY-9对松墨天牛3龄幼虫的LC_(50)分别为5.447×10^(6)孢子·mL^(-1)和8.733×10^(5)孢子·mL^(-1)。[结论]球孢白僵菌LCL-8和MLY-9对松墨天牛幼虫的致病力较高,可作为该害虫绿色防控的潜在生防资源。
[Objective]This study aimed to identify the pathogenic species and biological characteristics of two isolates of entomogenous fungi and evaluate their infection activity against Monochamus alternatus Hope larvae.[Method]Two isolates of entomopathogenic fungi isolated from different hosts and regions were purified and identified using morphological characteristics and sequence homology analysis of rDNA-ITS,TEF-1α,and Bloc regions.The spore diameter,colony growth rate,sporulation,spore germination rate,and biological activity against the 3rd instar larvae of M.alternatus were measured and compared.[Result]Both isolates were identified as Beauveria bassiana,designated as LCL-8 and MLY-9,respect-ively.The conidia of these isolates were spherical or nearly spherical,with diameters of(2.414±0.210)μm and(2.681±0.225)μm,respectively.Colony growth rates on PDA medium at 25℃were(4.050±0.052)mm·d^(-1) and(3.825±0.022)mm·d^(-1),and sporulation rates were(8.504±0.814)×10^(7) spores·cm^(-2) and(7.592±0.260)×10^(7) spores·cm^(-2),respectively.Spore germination rates at 24 h were(82.2±4.3)%for LCL-8 and(93.2±3.2)%for MLY-9.There were significant differences in spore diameters,colony growth rate and 24 h spore germination rate between the two strains(p<0.05).Both isolates showed high patho-genicity against the 3rd instar larvae of M.alternatus,with MLY-9 exhibiting higher virulence.At a concen-tration of 1×10^(8) spores·mL^(-1),the cumulative corrected mortality rates of LCL-8 and MLY-9 reached 100.0%on the 11th and 8th days post-inoculation,respectively,with LT50 values of 5.470 days and 5.335 days.Additionally,the LC_(50) values for LCL-8 and MLY-9 against the larvae were 5.447×10^(6) spores·mL^(-1) and 8.733×105 spores·mL^(-1),respectively.[Conclusion]Beauveira bassiana isolates LCL-8 and MLY-9 exhibit high pathogenicity against Monochamus alternatus larvae,making them promising biocontrol agents for the green prevention and management of this pest.
