摘要:嵩草属植物是青藏高原及其周边地区高寒草甸的建群种,分蘖能力强、根系发达,耐寒,是优良的牧草资源。目前对该类植物转录组情况的研究相对不足。该文采用Illumina HiSeq 2500高通量测序技术,对西藏嵩草(Kobresia tibetica)茎、叶、花及果实的混合样进行转录组分析。经拼接组装共获得63 837个Unigene,序列平均长度890.1bp,N50为1 342bp。将Unigene序列与5个主要的公共数据库(NR,Swiss-Prot,KEGG,GO,KOG)进行比对(Evalue<1e-5),共有40 705条Unigene获得了基因注释,占总Unigene的63.76%。通过KEGG pathways分析,共有10 043Unigene参与了274个代谢通路,获得了西藏嵩草分蘖和抗寒相关Unigene 179个。在15 044条Unigene中共搜索到5 310个SSR位点,其中二核苷酸和三核苷酸的重复类型占所有SSR位点的94.63%。这些数据为嵩草相关基因的发掘和SSR分子标记开发提供了重要依据。
Plants of genus Kobresiaserve as constructive species in alpine meadow systems of Tibet Pleatu and adjacent regions.Having high performance in spouting,proliferation and cold-resistance,they are widely considered as excellent grazing resource.However,the transcriptome information of Kobreasiais currentlly inadequate.In this study,the transcriptome of Kobresia tibetica is sequenced using Illumina Hiseq 2500 from a compounded sample of stem,leave and spikelet.In total,63 837 Unigene with an average length of 890.1bp and N50 of 1 342 bp are obtained by de novo assembly.AquiredUnigene sequences are compared with five main gene data base(NR,Swiss-Prot,KEGG,GO,KOG),40 705 Unigene have been annotated,63.76% of the total.10 043 Unigene are divided into 274 KEGG pathway.179 Unigene are found to be functional in spouting and cold resistance.Addtionally,5 310 SSR site are obtained from15 044 Unigene,of which 2base and 3base motifcompose 94.63% of total amount.These results will be helpful for the gene mining and molecular marker development in the future research.
