摘要:[目的]松材线虫引发的松树萎蔫病是世界性毁灭性病害,已在我国造成严重的生态与经济损失。早期、精准的诊断是实现有效防控的关键。本研究旨在通过比较蛋白质组学分析,筛选红松在松材线虫侵染过程中表达的差异蛋白,为诊断方法开发提供潜在靶标。[方法]以红松针叶为材料,设置接种松材线虫后0、1、5和20 d 4个时间点,开展定量蛋白质组学分析,结合生物信息学方法筛选上调表达的特异蛋白,并通过qRT-PCR验证其转录表达水平。[结果]共筛选出11种在侵染后持续显著上调的差异表达蛋白,进一步结合对照组无丰度、表达差异显著及功能结构域预测等标准,确定2种蛋白(DN8104_c0_g1_i1、DN14451_c0_g1_i1)具有作为早期诊断靶标的潜力。qRT-PCR验证结果与蛋白组学趋势一致。[结论]本研究鉴定的2种候选蛋白在红松受侵染后上调表达,且在未接种松材线虫的阴性对照中丰度为0,具备结构功能特征,具有作为松材线虫病早期诊断靶标的应用前景。研究为红松及其它松树树种的病害检测提供了理论基础与技术储备。
[Objective]Pine wilt disease,caused by pine wood nematode(Bursaphelenchus xylophilus),is a globally destructive disease,leading to serious ecological and economic losses in China.Early and accurate diagnosis is crucial for controlling pine wilt disease.This study aimed to screen the differentially expressed proteins in Pinus koraiensis during the infection by pine wood nematode through proteomics analysis,providing potential targets for the development of diagnostic methods.[Method]Needle tissues of P.koraiensis were sampled at four time points:0,1,5,and 20 days post-inoculation with B.xylophilus.Quantitative proteomics analysis was performed to screen the specific proteins that were up-regulated by bioinformatics methods,and their transcriptional expression levels were verified by qRT-PCR.[Result]A total of 11 differentially expressed proteins were found to be continuously and significantly upregulated after infection.Based on the criteria including absence in the control group,significant expression differences,and functional domain prediction,two candidate proteins(DN8104_c0_g1_i1 and DN14451_c0_g1_i1)were identified as early diagnostic targets.The qRT-PCR results were consistent with the proteomic expression trends.[Conclusion]The two candidate proteins identified in this study were up-regulated after infection of P.koraiensis,and the abundance was 0 in the negative control without inoculation of pine wood nematodes,which had structural and functional characteristics and had application prospects as early diagnostic targets for pine wilt disease.The research provides a theoretical basis and technical reserve for the disease detection of P.koraiensis and other pines.
