摘要:以5个品种(系)观赏凤梨的腋芽为外植体,研究了不同基因型、激素配比和苗龄对芽分化的影响。采用L9(34)正交试验分别针对TDZ,6-BA,NAA,IBA,蔗糖、大量元素及椰乳(CW)等因素进行优化,筛选出最佳的增殖培养基和生根培养基。结果表明:芽分化的最佳培养基为MS+TDZ 1.5 mg/L+6-BA 0.5mg/L。在此分化培养基上,紫星和红星的芽分化频率较高,达76.8%,70.6%。黄星、火炬和红剑芽分化频率较低,分别为44.2%,27.4%和24.9%。7 d苗龄的腋芽外植体最适于再分化。最佳增殖培养基为MS+TDZ 0.5 mg/L+6-BA 1.0 mg/L+NAA 0.2 mg/L+蔗糖40 g/L,其增殖系数达7.31。最佳的生根培养基为1/2 MS+NAA 0.5 mg/L+IBA 1.0 mg/L+CW 20 mL/L,生根率达60%以上。
Five ornamental bromeliads varieties were used to investigate the effects of different genotypes,hormone combinations and seedling ages on shoot differentiation.Based the recipe,the effects of TDZ,6-BA,NAA,IBA,sucrose,MS and coconut water(CW) were investigated to optimize the shoot regeneration system through L9(34) orthogonal experiment and to select the optimal rooting medium.The results showed that the medium optimal for adventitious buds was MS+TDZ 1.5 mg/L+6-BA 0.5 mg/L.In this differentiation medium,different ornamental bromeliads varieties differed greatly in their regeneration capacities,Zixing and Hongxing were 76.8% and 70.6%,respectively,but Huangxing,Huoju and Hongjian were 44.2%,27.4% and 24.9%,respectively.The explants for 7-day-old seedling had a high differentiation rate.The optimal medium for the propagation of adventitious buds consists of MS +TDZ 0.5 mg/L + 6-BA 1.0 mg/L +NAA 0.2 mg/L +sucrose 40 g/L,the multiplication coefficient reached to 7.31;and the optimal medium for the rooting was 1/2MS +NAA 0.5 mg/L + IBA 1.0 mg/L +coconut water extract 20 mL/L.Rooting rate was over 60%.
