摘要:目的采用响应面法优化大球盖菇中麦角硫因的提取工艺,并评价多种环境因素对其抗氧化能力的影响。方法本实验以大球盖菇次级菇为实验材料,首先确定提取方法,然后以温度、时间、料液比和乙醇浓度为单因素,考察它们对麦角硫因提取量的影响,在此基础上采用响应面法优化其提取条件,并且考察温度、pH和光照等外界环境因素对麦角硫因体外抗氧化能力稳定性的影响。结果选取溶剂提取法进行提取实验,确定最优提取条件为:提取温度70℃、乙醇浓度43%、提取时间32 min、料液比1:27(g/mL),在此条件下获得麦角硫因的最高提取量为(3.192±0.024)mg/g DW,与预测值接近;还发现麦角硫因的体外抗氧化能力受pH和紫外光照因素影响显著(P<0.05),受温度因素的影响不显著(P>0.05)。结论研究表明大球盖菇次级菇中麦角硫因含量较高,其体外抗氧化能力的稳定性易受光照、pH等因素影响。这为麦角硫因的后续开发利用提供数据支撑,也促进了大球盖菇次级菇的高值化利用。
Objective To optimize process conditions for the sub-quality Stropharia rugosoannulata ergothioneine extraction by response surface methodology and its stability of antioxidant activities.Methods After screening methods of extraction,the effects of temperature,time,concentration of ethanol and solid-liquid ratio on the extraction yield of ergothioneine in sub-quality Stropharia rugosoannulata were investigated using the single factor method.On the basis,the extraction process was optimized by response surface methodology,and the effects of temperature,pH and light on the stability of in vitro antioxidant capacity of ergothioneine were investigated.Results The solvent extraction method was used for the extraction,the optimum extraction conditions was confirmed as follows:Extraction temperature 70℃,concentration of ethanol 43%,extraction time 32 min,solid-liquid ratio 1:27(g/mL),the highest extraction yield of ergothioneine was(3.192±0.024)mg/g DW,which was close to the predicted value.Furthermore,the effect of pH and UV light on the in vitro antioxidant ability of the ergothioneine was significantly(P<0.05),and no significantly affect under the different temperature(P>0.05).Conclusion The results show that the content of ergothioneine in sub-quality Stropharia rugosoannulata is high,and the stability of its antioxidant capacity in vitro is easily affected by light,pH and other factors.This study provides data support for the subsequent development and utilization of ergothioneine,and also promotes the high-value utilization of sub-quality Stropharia rugosoannulata.
