摘要:为优化余甘子种子无菌萌发的激素条件以提高其发芽率,以赤霉素(GA_(3))和萘乙酸(NAA)为外源激素,分别设置3个浓度水平进行浸种催芽试验,系统分析不同浓度的GA_(3)和NAA对余甘子种子萌发及幼苗生长的影响。结果表明,GA_(3)浓度显著影响发芽指数,NAA浓度则显著影响发芽势。总体萌发效果以处理A_(2)B_(3)(1.0 mg/L GA_(3)+0.3 mg/L NAA)和处理A_(3)B_(1)(2.0 mg/L GA_(3)+0.1 mg/L NAA)最佳,发芽率均达到97.22%,发芽指数和发芽势分别较对照提高50%以上和1倍以上。幼苗生长动态分析表明,NAA主要影响发芽后21 d时的上胚轴长度,GA_(3)则显著影响35 d时的胚根长度。处理A_(2)B_(3)对余甘子幼苗生长的促进作用最为显著,其幼苗总长度在发芽后21 d和35 d时分别达5.36 cm和10.24 cm,较对照分别提高75.74%和87.55%。综上,1.0 mg/L GA_(3)+0.3 mg/L NAA为余甘子浸种催芽的最优激素组合,其种子无菌萌发及幼苗生长效果最佳。
To optimize the hormone conditions for the aseptic germination of Phyllanthus emblica seeds and increase their germination rate,gibberellin(GA_(3))and naphthalene-acetic acid(NAA)were used as exogenous hormones,and 3 concentration levels were set respectively for seed soaking and germination promotion experiments.The effects of different concentrations of GA_(3)and NAA on the germination of Phyllanthus emblica seeds and the growth of seedlings were systematically analyzed.The results showed that the concentration of GA_(3)significantly affected the germination index,while the concentration of NAA significantly affected the germination potential.The overall germination effect was best when treating A_(2)B_(3)(1.0 mg/L GA_(3)+0.3 mg/L NAA)and A_(3)B_(1)(2.0 mg/L GA_(3)+0.1 mg/L NAA).The germination rates of both treatments reached 97.22%,and the germination index and germination potential were increased by more than 50%and more than 100%respectively compared with the control.The dynamic analysis of seedling growth indicated that NAA mainly affected the length of the epicotyl at 21 days after germination,while GA_(3)significantly affected the length of the embryo root at 35 days.The treatment of A_(2)B_(3)had the most significant promoting effect on the growth of Phyllanthus emblica seedlings.The total length of the seedlings reached 5.36 cm and 10.24 cm at 21 days and 35 days after germination,respectively,which was 75.74%and 87.55%higher than that of the control.In conclusion,1.0 mg/L GA_(3)+0.3 mg/L NAA was the optimal hormone combination for seed soaking and germination promotion of Phyllanthus emblica,and it had the best effect on aseptic seed germination and seedling growth.
