摘要:【目的】验证弱毒真菌拟双色长喙壳(Ophiostoma pseudobicolor)接种日本落叶松(Larix kaempferi)后诱导其对强毒富士内生孢梗菌(Endoconidiophora fujiensis)抗性增强的生理机制,为林木病害防控提供新思路和新方法。【方法】以2年生日本落叶松为试验材料,通过人工接种方法评估喷施弱毒真菌孢子悬浮液诱导日本落叶松抗性增强水平;利用高效液相色谱和分光光度计法分析寄主防御酶(苯丙氨酸解氨酶PAL和过氧化物酶POD)和酚类化合物(原花青素、儿茶素、咖啡酸和花旗松素)在日本落叶松抵御强毒真菌中的作用。【结果】通过喷施拟双色长喙壳孢子悬浮液诱导的日本落叶松,在接种富士内生孢梗菌后,其韧皮部坏死斑面积显著小于喷施无菌水后接种富士内生孢梗菌的对照组坏死斑面积(在第28天小了75.43%);诱导后接种强毒真菌的日本落叶松韧皮部中的PAL、POD活性和4种酚类化合物含量均随时间延长呈不断上升趋势,在接种后第28天的2种酶活性和酚类化合物含量都显著高于未诱导处理和健康的日本落叶松。【结论】弱毒真菌拟双色长喙壳接种诱导能增强日本落叶松对强毒真菌富士内生孢梗菌的抗病性,且诱导抗病效果与韧皮部的防御酶活性和酚类化合物含量正相关。
【Objective】To verify the physiological resistance enhancement mechanism of Larix kaempferi to the hypervirulence Endoconidiophora fujiensis after inoculation with the hypovirulent Ophiostoma pseudobicolor,which provides new insights and methods for the prevention and control of forest diseases.【Method】Using two-year-old L.kaempferi as experimental subjects.The resistance enhancement level in L.kaempferi induced by spore suspension of O.pseudobicolor was evaluated through artificial inoculation method.The host defensive enzymes(phenylalanine ammonia-lyase PAL and peroxidase POD)and phenolic compounds(proanthocyanidins,catechins,caffeic acid and taxifolin)were analyzed using a high-performance liquid chromatography instrument and a spectrophotometer to investigate their roles in L.kaempferi resistance to E.fujiensis.【Result】The phloem necrotic lesion areas of L.kaempferi decreased by 75.43%after spraying the spore suspension of O.pseudobicolor and then inoculating with E.fujiensis compared with the treatment of spraying water and then inoculating with E.fujiensis after 28 d.The activities of PAL,POD and the contents of four phenolic compounds in the phloem of L.kaempferi showed a continuously increasing trend with the increase of time.After 28 days,the activities of both enzymes and the contents of phenolic compounds in the phloem were significantly higher than those in the unpretreatment and healthy L.kaempferi.【Conclusion】This study demonstrated that hypovirulent O.pseudobicolor induce resistance enhancement of L.kaempferi to the hypervirulence E.fujiensis,and the effect of induced disease resistance was related to the activity of defense enzymes and the content of phenolic compounds.
