摘要:为探究红花风铃木查尔酮合成酶(ChalconeSynthase,CHS)基因在类黄酮生物合成中的作用,从红花风铃木叶片中克隆HiCHS1基因,并利用生物信息学方法分析其序列特征和组织表达特异性,为红花风铃木类黄酮积累机制提供理论依据。结果表明:HiCHS1基因包含1个内含子和2个外显子,cDNA序列全长为279bp,编码93个氨基酸,蛋白分子量为10.1 kDa,理论等电点为8.96。该基因具有CHS基因家族典型功能结构域,无信号肽,不包含跨膜结构域。进化分析显示,HiCHS1与大花五桠果、巨桉、谷子、黄花蒿、决明的CHS蛋白亲缘关系较近,均含有保守基序motif1和motif2。启动子分析表明,HiCHS1可能受ABA诱导的胁迫响应调控。蛋白二级结构包含α-螺旋、β-折叠、无规则卷曲。此外,HiCHS1在不同组织中均有表达,在花和叶中表达量较高,在木质部中表达量最低。综上,HiCHS1为CHS家族非分泌蛋白基因,在细胞质中催化类黄酮生物合成,参与相关代谢通路。
To explore the role of the Chalcone Synthase(CHS)gene in flavonoid biosynthesis,the HiCHS1 gene was cloned from the leaves of Handroanthus impetiginosus,and its sequence characteristics and tissue-specific expression were analyzed using bioinformatics methods to provide a theoretical basis for the flavonoid accumulation mechanism in H.impetiginosus.The results showed that the HiCHS1 gene contains one intron and two exon regionswith a cDNA sequence of 279 bp,encoding 93 amino acids.The protein had a molecular weight of 10.1 kDa and a theoretical isoelectric point of 8.96.The gene exhibited the typical functional domain of the CHS gene family,lacked a signal peptide,and does not include transmembrane domains.Evolutionary analysis indicated that HiCHS1 was closely related to CHS proteins from Dillenia turbinate,Eucalyptus grandis,Setaria italica,Artemisia annua,Senna tora,and all contaied conserved motifs motif1 and motif2.Promoter analysis suggested that HiCHS1 was likely regulated by ABA-induced stress responses.The protein’s secondary structure consisted ofα-helices,β-sheets,and random coils.Furthermore,HiCHS1 was expressed in various tissues,with higher expression levels in flowers and leaves and the lowest in xylem.In conclusion,HiCHS1 was identified as a non-secretory CHS family gene that catalyzed flavonoid biosynthesis in the cytoplasm and participated in related metabolic pathways.
