摘要:The optimal basal medium and plant growth regulators (PGRs) combinations for the induction of adventitious roots were identified using leaves with petioles of the clone of Betula alnoides (coded as TC2) as explants. Subsequently, segments of the adventitious roots were employed to research the effects of their position, dark culture duration, and PGRs combinations on the callus and adventitious shoots induction. The optimal medium for leaf rooting was WPM supplemented with 0.1 mg l(-1) NAA, 0.02 mg l(-1) BA, with rooting rate reaching 100% and above six roots after 30-day culture. The basal root section was identified as the most suitable segment, and dark culture for 10 days was determined to be the optimal time for callus induction. The callus induction rate reached 100% on medium WPB5 supplemented with 1.0-1.2 mg l(-1) TDZ and 0.2 mg l(-1) NAA. The adventitious shoots induction rate of root calluses was more than 60% on medium WPM supplemented with 1.2 mg l(-1) BA and 0.5 mg l(-1) GA(3). The total proliferation coefficient exceeded 0.8. The optimal scheme yielded approximately 0.9 adventitious shoots from each root callus of B. alnoides.
