摘要:对多年生黑麦草Lolium perenne品种“德比”成熟种子的愈伤组织诱导和植株再生进行了探索研究。结果表明 :在附加 10mg/L 2 ,4 D ,5 0 0mg/LCH(水解酪蛋白 ) ,0 5mg/L 6 BA和 0 5mg/LNAA的N6培养基中愈伤诱导率最高 ,达 85 %。分化培养基以附加 1mg/L、6 BA、0 5mg/LKT的N6培养基为最优 ,分化率为 77 73%。在大量元素减半的N6培养基中附加 30g/L蔗糖及 0 2mg/LNAA进行生根培养 ,生根率达 10 0 %。
The objective of this study was to optimize tissue culture responses of turf-type perennial ryegrass (Lolium perenne L.) for genetic transformation, the effects of medium supplements on tissue culture responses were investigated with mature seeds of an elite cultivar, “Derby”, as explant. The best callus induction was obtained in N 6 medium with 10 mg/L 2,4-D, 500 mg/L CH, 0.5 mg/L 6-BA and 0.5 mg/L NAA, of which the induction rate was 85%. The highest differentiation rate, (77.13%), was achieved in the medium contained N 6 medium and 1 mg/L 6-BA, 0.5 mg/L KT. The root inducing medium, containing 0.2 mg/L NAA , 30 g/L Sucrose and N6 medium with half macro, gave 100% root inducing rate.
