摘要:桫椤(Alsophila spinulosa)是现存的唯一木本蕨类植物,是我国传统的中药材,富含内生菌资源。以桫椤新鲜茎干为研究对象,经内生菌液制备、培养基培养和纯化、菌落形态观察、DNA提取、PCR扩增和测序、NCBI数据库比对,对桫椤茎干的内生菌进行系统分离鉴定。从桫椤新鲜茎干中共分离鉴定到22种内生真菌,隶属于2门7纲9目15科16属,菌落颜色主要有黄色、黄褐色、白色和黑色等,菌落形态主要有毛毡状、绒毛状和蛛网状等;其中菌株AlS-F6、AlS-F15和AlS-F16的生长速率较其他菌株慢。从属水平看,木霉属(Trichoderma sp.)菌株的分离频率最高,为18.2%,是内生真菌的优势属。同时,从桫椤新鲜茎干中分离鉴定到26种内生细菌,隶属于3门4纲5目7科12属,从属水平看,泛东氏菌属(Pantoea sp.)的分离频率最高,为23.1%,是内生细菌的优势属。桫椤茎干中含有丰富的内生真菌和内生细菌,可为研究桫椤内生菌的共生关系及桫椤的药理作用奠定基础。
Alsophila spinulosa is the only extant arborescent fern and a traditional Chinese medicinal plant with abundant endophytic microbial resources.This study focuses on fresh A.spinulosa stems,employing methods such as endophytic fungal liquid preparation,medium cultivation,purification,strain morphological observation,DNA extraction,PCR amplification,sequencing,and NCBI database comparison to systematically isolate and identify endophytic fungi from the stems.A total of 22 species of endophytic fungi were isolated and identified from the fresh A.spinulosa stems.They belongs to 2 phyla,7 classes,9 orders,15 families,and 16 genera.Trichoderma sp.The colors of all strains were mainly yellow,yellow-brown,white and black.The strain morphologies were mainly felt-like,villous,and cobwebby.The growth rates of strains AlS-F6,AlS-F15 and AlS-F16 were slower than those of other strains.Trichoderma sp.was the most frequently isolated fungus,accounting for 18.2%of the total isolates,making it the dominant genus of endophytic fungi.A total of 26 endophytic bacteria were isolated and identified,belonging to 3 phyla,4 classes,5 orders,7 families,and 12 genera.Pantoea sp.was the most frequently isolated bacterium,accounting for 23.1%of the total isolates,thus making it the dominant genus of endophytic bacteria.The results of this study indicate that the stems of A.spinulosa contain a rich diversity of endophytic fungi and bacteria.This study provides a theoretical basis for further research on the symbiotic relationship between endophytic microbes and the medicinal properties of A.spinulosa.
