摘要:以马大杂种相思为研究对象,建立扦插基质简单、成分可控,不需特殊培养条件的瓶内扦插生根技术体系。为今后深入研究扦插生根机理,提供一种扦插基质成分及培养环境可控的技术平台,利于获得更准确、可靠的生根机理研究结果。8月采取马大杂种相思采穗圃的当年生腋芽饱满枝条为外植体,扦插基质由7 g/L琼脂和抑菌剂(多菌灵,百菌清)组成,筛选适宜的抑菌剂种类及浓度,结合对外植体类型和消毒方式的筛选,确定适宜的外植体类型及消毒方式;对IBA、NAA、ABT三种生长调节剂的浓度和处理时间进行研究,最终确定适宜的扦插生根处理。结果表明:两种抑菌剂的抑菌效果有显著差异,以百菌清为适宜的抑菌剂,浓度为0.2 g/L最佳;以中部带叶为最佳外植体,并以多菌灵0.8 g/L浸泡3 min对外植体进行预处理,并IBA 1.0 g/L处理2 h,在开放的实验台上接种至扦插基质中,生根率为100%。本研究在扦插基质中加入抑菌剂,不使用高温高压灭菌、超净工作台,建立了马大杂种相思开放式瓶内生根体系,对后续生根机理研究具有重要意义。
This study focused on the rooting of A. mangiumx A. auriculiformis under open condition, to establish an easy technique for rooting, it cultured explants in medium that was consisted several ingredients and composition-controllable, so that it could be helpful for studding the rooting mechanism. The shoots were gathered from one-year-old cutting orchard of A. mangiumx A. auriculiformis in August. The culture medium consisted of agar 7 g/L, and bacteriostats (Chlorothalonil or carbendazim). Chlorothalonil and carbendazim with different concentrations were added into medium, explant types and disinfection strategies were screened. The concentration and treatment time of IBA, NAA, ABT for rooting also studied. The results shown that, there were significant difference between chlorothalonil and carbendazim. Chlorothalonil was the optimal bacteriostat with concentration of 0.2 g/L. The 3-5th node with leaves were selected as optimal explant and immersed them on carbendazim for 3 minutes for pretreatment. Soaked explants into IBA 1.0 g/L for 2 h, and the highest rooting rate was 100%. A new cutting rooting method for A. mongiumx A. auriculiformis cutting was established, which cultured on an open condition without high temperature and pressure sterilization. This method could be useful for studying the rooting mechanism.
