摘要:【目的】明确食线虫真菌淡紫拟青霉T-DNA插入突变体几丁质酶和蛋白酶活性和致病力的改变。【方法】测定淡紫拟青霉T-DNA插入突变体几丁质酶和蛋白酶活性,及对南方根结线虫的致病力,分析它们与致病力的相关性。【结果】结果表明:14个淡紫拟青霉T-DNA插入突变体几丁质酶活力和蛋白酶活力由于T-DNA的插入均受到了不同程度的影响。对南方根结线虫卵的寄生率与原始菌株也存在分化,筛选获得了1株致病力增强的突变体BN-11和11株致病力明显下降的菌株;通过分析,这14个淡紫拟青霉T-DNA插入突变体几丁质酶和蛋白质酶的活性与寄生率之间不呈正相关性。【结论】获得了淡紫拟青霉T-DNA插入突变体几丁质酶和蛋白质酶的活性与致病力变化的证据,有助于淡紫拟青霉致病机制分析及致病相关基因的克隆。
【Objective】The objective of this study was to investigate that protease and chitinase and their pathogenicity against Meloidogyne incognita when the genome of mutants of nematophagous fungus Paecilomyces lilacinus are inserted by T-DNA.【Method】Chitinase and protease activity are important indicators which can evaluate the biological control potential of P.lilacinus.These chitinase activity and protease activity of the mutants were analyzed and pathogenicity against Meloidogyne incognita was assayed.【Results】The chitinase and protease activities of mutants were affected at different levels after T-DNA was inserted into genomes.The tested 14 mutants were different on parasitic rates of nematode eggs.The pathogenicity of 1 mutant was increased but The pathogenicity 11 mutants was decreased significantly.According to the analysis,no positive correlations were detected between the activities of protease,chitinase of mutants and parasitic rates.【Conclusion】The study confirmed that chitinase and protease activities and parasitic rates were affected at different levels when T-DNA inserted into genomes.Additionally,the study may be helpful to understand the pathogenic mechanism of P.lilacinus and clone genes related to pathogenicity.
