摘要:[目的]为研究侵染杨树的蚜虫可能传播的植物病毒病害害,控制蚜虫对于杨树生长的影响。[方法]本研究在1年生新疆杨叶片上接种白毛蚜,通过高通量测序与序列组装技术,采用系统发育分析与RT-PCR方法确定蚜虫可能传播的病毒种类。[结果]对转录组测序数据使用Trinity进行拼接组装,并与NCBI Genbank已知序列进行匹配,共发现793条可能的病毒碱基序列,47个可能的病毒种类,通过发育树比对与基因序列的同源性验证可知,侵染杨树的白毛蚜主要传播的病毒为苹果茎沟病毒与柑橘黄化脉明病毒。[结论]本研究采用一种不同于传统植物病毒鉴定、更加便捷的方法来对蚜虫可能传播的植物病毒病害进行研究,最终确定了白毛蚜在杨树体内主要传播的病毒种类,使用本方法有助于快速、准确的确定蚜虫-杨树互作体系中可能存在的病毒性植物病害,为杨树的病虫害防治提供了理论基础。
[Objective]This study aimed to investigate the potential transmission of plant viral pathogens by aphids to poplar trees and to control the impact of aphids on poplar trees.[Methods]Poplar leaf aphids(Chaitophorus populialbae Boyer de Fonscoloube)were inoculated on the leaves of one-year-old Populus alba Linn.var.pyramidalis.High-throughput sequencing and sequence assembly techniques were employed to analyze the viral sequences.Phylogenetic analysis and RT-PCR were used to identify the species of viruses that aphids may transmit.[Results]Transcriptome sequencing data were assembled using Trinity and blasted in the viral Ref-Seq of NCBI Genbank.A total of 793 nucleotide sequences potentially belonging to viruses and 47 possible virus species were identified.Phylogenetic analysis and homology comparison of gene sequences revealed that the dominant viruses transmitted by white aphids to poplar trees were apple stem grooving virus(ASGV)and citrus yellow vein clearing virus(CYVCV).[Conclusion]This study presents an efficient method,distinct from the traditional plant virus identification methods,providing a more convenient approach to investigating plant viral diseases potentially transmitted by aphids.The study ultimately identified the major virus species transmitted by white aphids to poplar trees.This method facilitates rapid and accurate determination of viral plant diseases in the aphid-poplar tree interaction system,laying a theoretical foundation for the prevention and control of diseases and pests in poplar trees.
