摘要:从新疆荒漠生长的一种白刺属植物茎韧皮部组织中分离到一株内生真菌(菌株编号:BCS-1)。该菌株不形成有性或无性繁殖结构,显微形态和培养性状与类丝核菌(Rhizoctonia-like fungi)十分相似,呈现形态各异的"念珠状"膨大细胞;基于ITS序列的BLAST搜索并没找到相似性高(≤90%)的匹配记录,然而LSU基因分析表明该菌株属于子囊菌盘菌目火丝菌科。查阅文献可知该菌株的培养性状与Tricharina属的一个无性型Ascorhizoctonia最为接近。以2份标准菌株(T.striispora和T.ochroleuca)为材料,利用LSU、ITS和RPB2为分子标记进行多基因系统发育分析,结果表明BCS-1菌株与Tricharina物种在同一进化分支(100%自展支持率),结合形态学特征将其鉴定为T.praecox(中国新记录种)。该研究提出了特殊无孢内生真菌鉴定的策略,对快速鉴定疑难标本和认识菌物生物多样性具有一定的参考价值。
Objective: An endophytic fungus BCS - 1 was isolated from the bark of an unidentified species of Nitraria, a desert plant in Xin- jiang. Lack of sexual or asexual fruiting structures made a challenge for having a proper identification. Method:Morphological and cultural characteristics of BCS - 1 were shown to be similar to some Rhizoctonia - like fungi, especially for monilioid cells that are highly variable in size and shape. Whereas the BLAST - search with ITS and LSU genes showed that this fungus belonged to the member of Pyronemataceae (Pezizales, Ascomycota), which made it to be assigned to Ascorhizoctonia, the anamorph of Tricharina. To further confirm identification, two authenticated type cultures of Tricharina ( T. striispora and T. ochroleuca) were used to perform a multi - locus sequence analysis based on LUS, ITS and RPB2 genes. Result: Three phylogenetie trees indicated that BCS - 1 formed a strongly supported clade with other Tricharina species (100% bootstrap value). Taken together i the evidence of combined morphological and molecular data is enough to place BCS - 1 strain as Ascorhizoctonia praecox, a new record species in China. Conclusion: This work presented a general strategy for identifying unique sterile endophytes, Which would aid in the identification of special specimen in a fast and accurate manner and increasing our knowledge a- bout the global fungal diversity.
