摘要:Arecanut(Areca cathecu L.) yellow disease is a destructive disease caused by the phytoplasma,a serious threat to the healthy development of arecanut industry in Hainan Province for a long time.The 16 S rRNA gene sequence was employed as the target to establish a rapid and efficient detection system for arecanut yellow leaf phytoplasma belonging to 16 SrI group in China by loop mediated isothermal amplification(LAMP) in the study,with testing the specificity,sensitivity and stability of the detection system.LAMP technology aiming to 16 SrI group arecanut yellow leaf phytoplasma in China was used to detect the samples of the arecanut yellow leaf disease from different areas of Hainan province.The results showed that two sets of LAMP detection primers,16 SrDNA-2 and 16 SrDNA-3 were obtained for the 16 SrI group arecanut yellow leaf phytoplasma in China based on the 16 S rRNA gene sequence,both with good specificity and stability;The lowest detection limit of sensitivity of the two sets of LAMP detection systems was the same,200 ag/μL;Phytoplasmas were all detected in arecanut yellow leaf disease samples from Baoting,Tunchang,Wanning in Hainan province of China by using the two sets of LAMP primers 16 SrDNA-2 and 16 SrDNA-3,and no phytoplasma was detected in the negative control.LAMP detection technology is fast,efficient,easily operate and the results are visual,suitable for application and research in the rapid diagnosis of arecanut yellow disease in the field,the detection of seedlings with pathogen and the breeding of resistant arecanut varieties.It is of great significance for the detection and identification of the arecanut yellow disease pathogen,the spread and scientific control of the disease.
