摘要:火力楠具有重要经济价值与生态功能,但缺乏其基因组信息,限制了其分子生物学、基因功能与分子育种的相关研究。以火力楠根、茎和叶为材料,利用Illumina高通量测序技术进行转录组测序,并对得到的测序数据进行de novo组装,获得97 503条Unigenes,N50为1 010 bp、平均长度852 bp。与公共数据库进行比对,注释到NR、Swiss-Prot数据库的Unigenes分别为60 926、44 249条。将Unigenes与COG数据库比对,有42 344条Unigenes成功注释,根据功能大致分成25类;与GO数据库比对,有15 947条Unigenes获得注释,按功能可分为生物学过程、细胞组分和分子功能3大类55亚类,其中参与的生物学过程较多;以KEGG数据库参考,有26 267条Unigenes参与330条代谢途径分支,以代谢相关的途径较为集中;差异基因表达分析我们得出,根与茎的差异表达基因为2 826个,根与叶之间的差异表达基因为3 230个,这两组差异基因相对校多,除此之外茎与叶差异基因相对较少;差异基因的GO分类、GO富集以及pathway富集分析表明,主要在细胞组分、蛋白质活性、生物合成以及代谢过程等所占比例较高。这些研究结果极大地扩充了火力楠的基因资源,将有助于火力楠基因的发掘与利用、分子标记的开发及其种质资源遗传改良的研究等。
Michelia macclurei is one of important economic value and ecological function tree. However, genomic information of this specie has been lack, leading to limited research progress in molecular biology, gene functions and molecular breeding. We have sampled the roots, stems and leavesof M. macclurei to conduct a transcriptomic analysis using the Illumina Hi Seq 2000 technical platform. In total 97 503 unigenes with an average length of 852 bp and N50 of 1010 bp were obtained by de novo assembly. For the Unigenes, 60 926 and 44 249 Unigenes had significant similarity with known data bank in NR, Swiss-Prot, respectively. The 42 344 unigenes were annotated in clusters of orthologous groups of proteins(COG) and assigned into 25 clusters. The 15 947 unigenes were annotated in gene ontology(GO) and grouped into biological processes,cellular components and molecular function three functional categories, 55 sub-categories. The biological processes were most commonly existed.A total of 26 267 Unigenes were divided into 330 KEGG pathways whose functions focused on metabolism. The differentially expressedgenes analysis showed that there were more genes between the root and stem, root and leaf, which were 2 826, 3 230,respectively, compared with less differentially expressed genes between the stem and leaf. GO classification,enrichment and pathway enrichment analysis showed that differentially expressed genes were mainly higher proportion in cellular component, protein activity, biosynthesis and metabolism. The sequence data of M. macclurei will be helpful to the gene discovery and utilization, molecular marker development and genetic improvement for this specie in future.
