摘要:利用8对SSR荧光标记对甘肃武都油橄榄品种园收集保存的113份品种资源进行遗传检测,将PCR扩增测序结果与油橄榄品种SSR共享数据库WOGBC和OD进行比对,鉴别油橄榄品种资源。结果表明,相同品种同一SSR位点等位基因数值在两个数据库中普遍存在差异,综合分析将纵向差值A和B频率≥30以及横向差值C频率≥10%作为可信对照,基于此将113份油橄榄品种SSR检测数据与两个数据库共有品种数据进行对比,共鉴别出油橄榄品种26个,成功率仅为23.0%,因此,检测结果稳定、适于油橄榄品种准确鉴定的遗传标记有待进一步开发。
Genetic analysis of 113 olive cultivars collected and preserved in Wudu olive cultivar orchard in Gansu Province was performed by using 8 pairs of fluorescence-labelled SSR markers.The PCR amplification sequencing results were compared with the shared SSR database WOGBC and OD to identify olive cultivars.The same SSR locus values of the same cultivar were widely different in the WOGBC and OD databases.Comprehensive analysis results took the frequency of longitudinal difference value A and B greater than or equal to 30%and the frequency of lateral difference value C greater than or equal to 10%as a trusted control.Then SSR test data of 113 olive cultivars were compared with the data of the two databases,and 26 olive cultivars were identified.The success rate was only 23.0%.Therefore,genetic markers with stable detection results and accurate identification of olive cultivars need further development.
