摘要:采用随机扩增多态性DNA(RAPD)分子标记技术对5个崖柏(Thuja sutchuenensis)天然居群的遗传变异进行研究。7个引物共检测44个位点,其中多态位点34个,占77.27%。与其它濒危植物相比,崖柏种内遗传多样性较为丰富。但居群间遗传分化占到33.68%。居群间遗传距离与空间距离不相关。根据崖柏居群遗传特征,初步探讨保护策略。
RAPD molecular markers were used to assess genetic variation within and among five natural populations of Thuja sutchuenensis (Cupressaceae). Seven random primers detected 44 loci of which 34 (77.27) were polymorphic. Compared with other endangered plants, the total genetic diversity of T. sutchuenensis was relative high. But the coefficient gene differentiation (Gst) among populations was up to 0. 336 8. The genetic distances were also caculated, and did not correspond to geographic distance. The strategies of conservation for T. sutchuenensis based on genetic diversity analysis were proposed finally.
