摘要:我国重要板栗品种‘燕山早丰’常年受到复合型种实腐烂病的危害,而其病原组成和致病性至今不清。通过科赫法则,采用分子系统发育分析方法结合形态学特征,分离鉴定了病原菌,并对病菌分生孢子、板栗内生菌致病性及温度对致病性的影响做了测定。结果表明,‘燕山早丰’种实腐烂病的病原菌分布在葡萄座腔菌属(Botryosphaeria)、间座壳属(Diaporthe)、镰刀菌属(Fusarium)、单端孢属(Trichothecium)、刺盘孢属(Colletotrichum)、链格孢属(Alternaria)及毛二孢属(Lasiodiplodia)中,而以葡萄座腔菌(B.dothidea)、桂冠间座壳菌(Diaporthenobilis)和毛二孢为主要致病种类。低温能显著影响病菌的致病性并减少致病菌种类,病菌分生孢子具致病性,‘燕山早丰’种仁的内生真菌间座壳菌和链格孢菌的菌株可表现致病性。研究首次系统揭示了一种板栗品种种实腐烂病的病原结构及其致病性,为针对性地探索和建立有效防治策略和技术奠定了基础。
Castanea mollissima var.Yanshanzaofeng,a significant chestnut variety in China,is perennially affected by a complex fruit/seed rot disease,whose pathogenic composition and virulence have remained unclear.In this study,Koch's postulates were employed to isolate and identify the causative pathogens,combining molec-ular phylogenetic analysis with morphological characteristics.Furthermore,the pathogenicity of fungal conidia,the virulence of endophytic fungi within chestnuts,and the impact of temperature on pathogenicity were evaluated.The results indicated that the pathogens responsible for fruit/seed rot in‘Yanshanzaofeng’chestnuts were distrib-uted across several genera,including Botryosphaeria,Diaporthe,Fusarium,Trichothecium,Colletotrichum,Al-ternaria and Lasiodiplodia.Among these,B.dothidea,D.nobilis and Lasiodiplodia species were identified as the primary pathogenic species.It was found that lower temperatures significantly affected the virulence of these pathogens and reduced the diversity of pathogenic species.The conidia of these fungi were determined to be pathogenic,and strains of Diaporthe and Alternaria endophytic fungi within the chestnut also exhibited patho-genicity.This study systematically revealed,for the first time,the pathogenic structure and virulence of a fruit/seed rot disease in a chestnut variety,which lay a crucial foundation for potential exploration and the estab-lishment of effective prevention and control strategies and techniques.
