摘要:【目的】木姜叶柯茶药历史悠久,嫩叶中含有丰富的黄酮类、根皮苷等成分,因其对于糖尿病等治疗疗效显著而备受关注。基于引种是扩大资源的重要方式,本研究拟利用多个统计方法分析木姜叶柯不同种源引种到多个异质园基地后主要药用成分的稳定性及变异特征,为其引种及资源利用奠定基础。【方法】将6个省区12个种源分别引种到3个异质园试验基地,测定药效成分总黄酮、三叶苷、根皮苷、二氢查耳酮、三叶苷/根皮苷比值,利用单因素方差分析(one-way ANOVA)、多重比较(Duncan’s)、主成分分析和聚类分析4种统计方法比较和分析引种前后主要药用成分的稳定性、差异、综合得分及聚类特点。【结果】发现引种后木姜叶柯的主要药效成分相似度为0.875~1.000,证实引种后主要药效成分相对较稳定。本试验共引种36次,研究获知总黄酮、三叶苷、根皮苷、二氢查耳酮、三叶苷/根皮苷比值等高于原产地时引种次数分别为21、10、27、14和5,占总次数的58.33%、27.78%、75.00%、38.89%和13.89%。聚类分析发现在欧氏距离为5时36份试验材料聚成4类,其中第3类包括11份,其总黄酮、三叶苷、根皮苷和二氢查耳酮含量均较高,大多高于原产地。主成分分析发现浙江江山、湖南溆浦、广东惠州、江西赣州、广西巴马、湖南芷江等8个种源综合排名靠前,均在聚类分析的第3类中。主成分分析和聚类分析表明浙江江山、广西巴马、广东惠州及江西的赣州、德兴和婺源6个种源,浙江江山、湖南溆浦、江西的赣州和横峰4个种源,湖南溆浦、广东惠州、广西巴马和江西德兴4个种源,分别在浙江富阳、浙江江山和湖南溆浦表现较好,且各主要药效成分含量基本高于均值。【结论】浙江江山、湖南溆浦、广东惠州、江西赣州、广西巴马、湖南芷江等8个种源在异质园基地综合表现均较好,不仅药效成分稳定,且部分成分含量超过原产地,建议引种时重点考虑。
[Objective]The tea medicine of Lithocarpus litseifolius has a long history.The young leaves are rich in flavonoids,phloridzin and other ingredients.It has attracted much attention because of its significant curative effect on diabetes and other diseases.Based on the fact that introduction is an important way to expand resources,this study intends to use multiple statistical methods to analyze the stability and variation characteristics of the main medicinal components after the introduction of different provenances to multiple heterogeneous garden bases,thus laying a foundation for its introduction and resource utilization.[Method]In this paper,twelve provenances from six provinces were introduced to three heterogeneous garden experimental bases.The total flavonoids,trilobatin,phlorizin,dihydrochalcone and the ratio of trilobatin/phlorizin were determined.Four statistical methods including one-way analysis of variance(one-way ANOVE),multiple comparisons(Duncan's),principal component analysis and cluster analysis are used to compare and analyze the stability,differences,comprehensive scores and cluster characteristics of major medicinal components before and after introduction of Lithocarpus litseifolius.[Result]The study found that the main components similarity of Lithocarpus litseifolius were 0.875-1.000,which confirmed that the main components were relatively stable after introduction.A total of 36 introductions were conducted in this experiment,and the number of introductions of total flavonoids,trilophyls,phlorizin,dihydrochalcone and trilophyllin/phlorizin ratios higher than those in the place of origin were 21,10,27,14 and 5,accounting for 58.33%,27.78%,75.00%,38.89% and 13.89% of the total number of introductions,respectively.Cluster analysis showed that 36test materials were clustered into 4 categories when the Euclidean distance was 5,of which 11 were in the third category.The contents of total flavonoids,trilobatin,phloridzin and dihydrochalcone were higher,mostly higher than those in the origin.In the principal component analysis,8 provenances,including Jiangshan City of Zhejiang Province,Xupu County of Hunan Province,Huizhou City of Guangdong Province,Ganzhou City of Jiangxi Province,Bama County of the Guangxi Zhuang Autonomous Region,Zhijiang County of Hunan Province were ranked in the top of the comprehensive ranking,all in the third category of cluster analysis.Principal component analysis and cluster analysis showed that the six provenances of Jiangshan in Zhejiang,Bama in Guangxi,Huizhou in Guangdong and Ganzhou,Dexing and Wuyuan in Jiangxi,the four provenances of Jiangshan in Zhejiang,Xupu in Hunan,Ganzhou and Hengfeng in Jiangxi,and the four provenances of Xupu in Hunan,Huizhou in Guangdong,Bama in Guangxi and Dexing in Jiangxi performed well in Fuyang in Zhejiang,Jiangshan in Zhejiang and Xupu in Hunan,respectively,and the content of each main pharmacodynamic component was basically higher than the mean value.[Conclusion]Eight provenances,including Jiangshan of Zhejiang,Xupu of Hunan,Huizhou of Guangdong,Ganzhou of Jiangxi,Bama of Guangxi,Zhijiang of Hunan,ect.all had good comprehensive performance in the heterogeneous garden base,not only the pharmacodynamic components were stable,but also the content of some components exceeded the place of origin,which is recommended for introduction.
