摘要:[目的]系统解析核桃JrWOX4b基因的过表达对‘84K’杨不定根、茎和叶片生长发育的影响,并对其分子机制进行初步探索。[方法]以前期创制JrWOX4b基因异源过表达‘84K’株系和野生型植株为材料,分析JrWOX4b基因的过表达对‘84K’杨不定根、茎和叶片的表型和组织结构的影响,通过转录组测序和生物信息学技术进行基因表达模式的分析。[结果]表型数据分析结果显示,JrWOX4b基因的过表达显著改变转基因‘84K’杨根系构型:总根长和总根面积等显著增加。JrWOX4b基因的过表达显著抑制了转基因株高和地径生长,且叶片明显向近轴面卷曲;组织学研究结果显示,JrWOX4b过表达株系中形成层细胞的层数显著增加。叶片主脉中维管束占叶脉的比例增大,且韧皮部和木质部细胞排列不规则;转录组测序和生物信息学分析共获得‘84K’杨和JrWOX4b基因过表达株系茎中显著差异表达基因1 535个。GO富集分析结果显示,茎段中差异表达基因主要富集在多细胞器官发育、发育过程和组织结构发育等功能中。KEGG富集分析结果表明,茎段中差异表达基因主要富集在苯丙氨酸、酪氨酸和色氨酸的生物合成,戊糖和葡萄糖醛酸盐相互转化和苯丙胺代谢等通路中。[结论]JrWOX4b基因过表达显著增强‘84K’杨树不定根的根系发育,减小其株高,并使其叶片向近轴卷曲。初步分析了转基因植株茎中差异表达基因富集的代谢通路,为进一步揭示JrWOX4b基因在根系、茎和叶片发育过程中的分子调控机制奠定基础,同时也为利用分子生物学手段精确育种提供优质基因资源。
[Purpose]To systematically investigate the effects of overexpressing the walnut JrWOX4b gene on adventitious root formation,stem,and leaf development in Populus‘84K’,and to preliminarily explore the underlying molecular mechanisms.[Methods]Previously generated JrWOX4b heterologous overexpression transgenic‘84K’poplar lines and wild-type plants were used.Phenotypic traits and tissue structures of adventitious roots,stems,and leaves were analyzed.Transcriptome sequencing and bioinformatic analyses were employed to study gene-expression patterns.[Results]Phenotypic data revealed that overexpression of JrWOX4b significantly altered the root architecture of transgenic‘84K’poplar,notably increasing total root length and total root surface area.Conversely,plant height and basal stem diameter were significantly reduced,and leaves exhibited pronounced adaxial curling.Histological observations revealed a marked increase in cambium cell layers in transgenic lines.In leaf main veins,the proportion of vascular bundles increased,accompanied by irregular organization of both phloem and xylem cells.RNAseq identified 1,535 differentially expressed genes(DEGs)in stems between transgenic and wild-type plants.GO enrichment analysis revealed that DEGs were mainly associated with multicellular organ development,developmental processes,and tissue structure development.KEGG enrichment analysis indicated that DEGs were significantly enriched in pathways including phenylalanine,tyrosine,and tryptophan,interconversion of pentose and glucuronate,and phenethylamine metabolism.[Conclusion]Overexpression of JrWOX4b significantly promotes adventitious root development in Populus of‘84K’,reduces plant height,and induces adaxial leaf curling.Preliminary analysis of the enriched metabolic pathways of DEGs in stems provides insights into the molecular regulatory mechanisms of JrWOX4b in root,stem,and leaf development,and offers valuable genetic resources for precision breeding through molecular approaches.
