摘要:[目的]探明侧柏古树扦插繁殖困难的原因及生理反应,为保护和繁殖侧柏提供科学依据。[方法]以不同树龄(5、100、700年生)侧柏插穗为对象,研究扦插生根情况,测定机械损伤处理后侧柏古树插穗的生根时间、生根率和生根数,同时对不同生根阶段和不同树龄的侧柏扦插插穗进行生理生化指标测定。[结果]1)100年生和700年生侧柏母树插穗的生根率和生根数显著低于5年生插穗(p<0.05)。2)解剖学观察到100年生和700年生侧柏母树插穗的愈伤组织木质素沉积。3)与未机械损伤处理(或对照)相比,机械损伤处理能显著提高100年生和700年生母树插穗的生根率(p<0.05)。4)机械损伤处理后的不同树龄母树插穗中可溶性糖、淀粉、H_(2)O_(2)含量和SOD、POD活性均显著高于未机械损伤处理的插穗,机械损伤的插穗在不定根形成阶段CAT活性、VB_(6)含量显著高于未机械损伤插穗(p<0.05)。机械损伤处理后,100年生和700年生插穗中可溶性糖、淀粉含量和SOD、POD、CAT活性低于5年生插穗,MDA、H_(2)O_(2)含量和ROS产生速率则显著高于5年生插穗(p<0.05)。[结论]古树插穗较幼龄插穗生根能力差,愈伤组织木质化是阻碍古树插穗不定根形成的重要因素;机械损伤能够有效突破木质化的愈伤组织,增强古树及难生根树种的扦插再生能力。
[Objective]To investigate the reasons for the difficulty of propagation of ancient Platycladus orientalis cuttings and their physiological responses,providing a scientific basis for the conservation and propagation of P.orientalis.[Methods]The rooting of P.orientalis cuttings of different ages(5,100 and 700 years old)was investigated.Rooting time,rooting rate and rooting number of P.orientalis cuttings were determined following mechanical wounding treatment.Additionally,physiological and biochemical indices of P.orientalis cuttings at different rooting stages and different ages of trees were measured.[Results](1)The rooting rate and number of roots in cutting from 100-and 700-year-old P.orientalis mother trees were significantly lower than those from 5-year old trees(p<0.05).(2)Anatomical observations revealed increased lignin deposition in the callus of 100-and 700-year-old P.orientalis mother trees cuttings.(3)Compared with the non-mechanical wounding treatment(or control),the mechanical wounding treatment significantly increased the rooting rate in cuttings from 100-and 700-year-old P.orientalis mother trees(p<0.05).(4)Soluble sugars,starch,H_(2)O_(2) content,SOD and POD activities were significantly higher in mechanically wounded cuttings of mother trees of different ages than those of unmechanically wounded cuttings.CAT activity and VB_(6) content were significantly higher in mechanically wounded cuttings than those of unmechanically wounded cuttings at the stage of adventitious root formation(p<0.05).After mechanical wounding treatment,the soluble sugar,starch content and activities of SOD,POD and CAT in 100-and 700-year-old cuttings were lower than those in 5-year-old cuttings,while MDA,H_(2)O_(2) content and ROS production rate were significantly elevated(p<0.05).[Conclusion]The rooting ability of ancient tree cuttings is inferior to that of young cuttings.Lignification of the callus plays an important role in hindering adventitious root information in ancient tree cuttings.Mechanical wounding can effectively break through the lignified callus,and enhance the regeneration ability of cuttings of ancient trees and difficult-to-root tree species.
