摘要:针对在土沉香Aquilaria sinensis(Lour.)Gilg组织培养过程中外植体的有效消毒方式问题,以1年生土沉香实生苗为材料,开展了茎段外植体次氯酸钠(5%、10%、20%)和升汞(0.05%、0.1%、0.2%)3个浓度水平的消毒试验;在此基础上,开展了外植体(叶片、顶芽和茎段),次氯酸钠消毒时间(0,3,5 min)和升汞消毒时间(0,3,5 min)的3因素3水平正交L9(34)组合排列试验。结果表明:茎段外植体采用10%浓度的次氯酸钠、0.1%浓度的升汞消毒,效果较好;茎段较叶片、顶芽的污染率和死亡率低,存活率高,为适宜的外植体。最佳消毒方式为75%的酒精浸泡30 s,0.1%升汞消毒5 min,该方法污染率最低、存活率最高,分别为43.33%、31.67%。
Focused on the issues of explants' effective disinfection schemes in the tissue culture of Aquilaria sinensis,one-year-old seedings of Aquilaria sinensis in the nursery were used as propagation material.Using stem explants,two disinfection experiments with three concentrations of NaClO(5%,10% and 20%) and HgCl2(0.05%,0.1% and 0.2%) were carried out.Based on the result,an orthogonal experiment with L9(34) array of three factors and three levels,namely explants(leaf,terminal bud and stem),NaClO's disinfection time(0,3,5 min) and HgCl2's disinfection time(0,3,5 min),was conducted.The results show the optimal concentration of NaClO and HgCl2 was 10% NaClO and 0.1% HgCl2 respectively.Compared with the explants of leaf and terminal bud,stem explants were much better for disinfection in view of its lower rate of contamination and mortality and relatively higher survive rate.The optimal disinfection schemes was that the stem explants were immersed in alcohol(75%) for 30 seconds,and then were immersed in HgCl2(0.1%) for 5 minutes with the lowest contamination rate(43.33%) and the highest survive rate(31.67%).
