摘要:以“纸皮”扁桃为试材,筛选水酶法适宜的酶,进而在单因素(料液比、酶添加量、pH、酶解时间、酶解温度)实验基础上,利用Box-Behnken响应面优化获得最佳提取工艺。结果表明,水酶法制备“纸皮”扁桃油的最佳工艺为:酶为果胶酶和水解蛋白酶的复合酶(果胶酶活性∶水解蛋白酶活性=1∶5),酶解pH 3.81,料液比1∶5.135,酶添加量为果胶酶∶水解蛋白酶/g样品=237.5 U∶1187.6 U/g样品,酶解温度47.59℃,酶解时间3 h,出油率为60.73%。
Aqueous enzymatic extraction(AEE)of oil from“Zhipi”,the trial material,almond kernels was performed.Firstly,the suitable enzyme was screened.After that,the box Behnken response surface was used to optimize the extraction process on the basis of single factor(liquid/solid ratio,mixed enzyme amount,time,and temperature)test.The results showed that the optimal extraction conditions for“Zhipi”almond oil yield were mixed enzyme as pectinase activity:hydrolytic protease activity=1∶5,pH as 3.81,liquid/solid ratio as 1∶5.135,mixed enzyme amount as 237.5 U∶1187.6 U/g,temperature as 47.59℃.Under the optimum conditions,the experimental oil extraction yield was 60.73%.
