摘要:对农杆菌EHA105转化柚木愈伤组织的适宜条件进行了优化。EHA105转化柚木愈伤组织的适宜预培养时间为11-13 d,菌液光密度D600 nm为0.4-0.5,共培养时间为7-9 d,共培养的pH为6.0-6.2。EHA105共培养对柚木愈伤组织有一定的毒害作用,共培养时间超过10d,可导致愈伤组织和茎段外植体的严重死亡。不同无性系对EHA105侵染的敏感性不一样,选择适宜的无性系可显著提高转化效率。
The genetic transformation of teak callus mediated by Agrobacterium tumefeciens strain EHA105 was discussed. The optimum pre-eulture time for callus induction of stem section explants is 11 to 13 days, D600sm value of bacterium inoculum solution is between 0. 4 to 0. 5, co-culture time is 7 to 9 days and pH value of co-culture medium is 6. 0 to 6.2. The infection of EHA105 is toxic to explants and there will be high death rate of explants if the co-culture time is longer than 10 days. Different clones have different susceptibility to the infection of EHA105 and clones with high susceptibility should be selected so that the transformation efficiency of teak can be greatly improved .
