摘要:弗栎象甲是危害弗吉尼亚栎的重要种实害虫。为了寻找对弗栎象甲具有高致病性的昆虫病原微生物,该研究采用组织分离法对罹病弗栎象甲病原微生物进行分离,通过形态学并结合分子生物学方法对病原微生物进行物种鉴定,采用扩增子测序法分析病原细菌的来源,采用生物测定法评价病原微生物的致病性。从弗栎象甲体内分离到病原细菌和病原真菌各1种。综合分析分离菌株的形态特征、培养性状和rDNA序列,确定病原细菌为粘质沙雷氏菌,病原真菌为球孢白僵菌。扩增子分析发现,粘质沙雷氏菌仅以低丰度存在于弗栎象甲幼虫体内。生物测定结果表明,弗栎象甲幼虫取食获得FL21菌株后6 d的半数致死浓度LC_(50)为4.71 cfu/mL;而采用浸渍法对弗栎象甲幼虫接种FLBJJ4菌株,接种后13 d的半数致死浓度LC_(50)为1.40×10~3孢子/mL。分离的粘质沙雷氏菌和球孢白僵菌对弗栎象甲幼虫均具有较高的致病性,开发利用价值较高,有望应用于弗栎象甲的生物防治。
Curculio victoriensis is an important acorn pest of Quercus virginiana.To identify entomopathogenic microorganisms with high pathogenicity to C.victoriensis,we isolated the entomopathogenic microorganisms from the diseased acorn weevils by tissue isolation approach,identified the species of the entomopathogenic microorganisms through morphology in combination with molecular biology,analyzed the source of the entomopathogenic bacteria using amplicon sequencing,and evaluated the pathogenicity of the isolated entomopathogenic microorganisms by bioassay.In the study,one isolate of entomopathogenic bacteria and 1 isolate of entomopathogenic fungi were got from the diseased acorn weevils.Based on morphological characteristics,cultural traits,and rDNA sequences,the isolated entomopathogenic bacterium was identified as Serratia marcescens and the isolated entomopathogenic fungus was identified as Beauveria bassiana.Amplicon sequencing showed that S.marcescens existed in the larvae of the acorn weevil with low abundance.The bioassay results showed that the LC 50 value of the S.marcescens FL21 isolate was 4.71 cfu/mL,measured 6 days after infecting C.victoriensis larvae with S.marcescens.The LC 50 value of the B.bassiana FLBJJ4 isolate was 1.40×103 conidia/mL at 13 days after the larvae of C.victoriensis were infected with B.bassiana.S.marcescens and B.bassiana isolated in this study exhibit high pathogenicity to the larvae of C.victoriensis,which have high exploitation value and are expected to be applied in the biological control of C.victoriensis.
