摘要:以优株无性系继代组培苗(JXFY1和ZJLS13)为试验材料,采用单因素和双因素试验设计,研究不同浓度的3种植物生长调节剂(ABT,IAA,NAA),DCR和1/2MS 2种培养基类型及河沙、轻基质和土壤3种培养基支撑物等对2种基因型沙氏鹿茸草组培苗生根的影响。结果表明,培养基中添加IAA的JXFY1组培苗生根率以及根系形态均处于较高水平,平均生根条数、平均最长根长和生根率均达到较大值,分别为3.06条/株、2.13 cm和59.36%,但其生根率和平均生根条数在不同植物生长调节剂水平间差异不显著(P=0.991和P=0.307),而ABT处理显著提高了ZJLS13的生根率(P=0.048),其中以1.0 mg/L处理的生根率及根系形态均最佳,平均生根条数、平均最长根长和生根率分别为2.91条/株、1.35 cm和64.48%;2种基因型在DCR培养基上的生根率均极显著高于1/2MS培养基,但基本培养基对不定根诱导效率的影响随激素组合而存在差异;在沙壤土、轻基质中试管苗生根率与对照差异不显著,轻基质中生根的组培苗移栽成活率却显著降低(P<0.001)。总体来看,DCR培养基生根效果最佳,沙壤土和琼脂均可作为培养基支撑物,1.0 mg/L IAA或IBA有利于JXFY1组培苗生根,1.0 mg/L ABT适宜ZJLS13组培苗生根。
Two different genotypes of shoot sections of superior clones(JXFY1 and ZJLS13)of M.savatieri were used as materials.The effect of factors such as PGRs,genotypes,baseal medium and medium supportings were studied for rooting frequency using single factor or two factors experimental design.The effects of different types and concentrations of PGRs on rooting parameters of M.savatieri,i.e.rooting rate,number of adventitious roots,average longest root length,were investigated.Results indicated that,on the medium with IAA,the rooting rate and root morphology of JXFY1 were all at a high level,the average number of rooting,the average longest root length and the rooting rate were 3.06,2.13 cm and 59.36%,respectively.However,there was no significant difference in the rooting rate and the average rooting number among different PGRs(P=0.991 and P=0.307).ABT treatment significantly increased the rooting rate of ZJLS13(P=0.048),when on the medium with 1.0 mg/L ABT,the root had the best quality,the average rooting number was 2.91,the average longest root length was 1.35 cm and rooting rate was 64.48%.The rooting rate of two genotypes on DCR medium was significantly higher than that on 1/2MS medium,but the effect of basic medium on adventitious root induction efficiency was different with PGRs combination;The rooting rate of seedlings in soil and light medium was not significantly different from that of control,but the survival rate of tissue culture seedlings rooting in light medium was significantly decreased(P<0.001).In general,DCR medium had the best rooting efficiency,soil and agar could be used as medium supporting.1.0 mg/L IAA or IBA was favorable for rooting of JXFY1 seedlings,and 1.0 mg/L ABT was suitable for rooting of ZJLS13.
