摘要:以轻木种子发芽后的芽苗为外植体材料,研究不同激素配比的培养基对其丛生芽增殖及生根培养的影响,筛选适宜各阶段培养的最佳培养基配方。结果表明:轻木种子A用0.1%的HgCl2处理15 min消毒效果最好,污染率仅为6.6%。芽苗在S4增殖培养基(MS+6-BA0.5 mg·L^(-1)+NAA0.2 mg·L^(-1))中生长最佳,增殖效果最好;T2生根培养基(1/2MS+IBA0.3 mg·L^(-1)+NAA0.1 mg·L^(-1))生根率可达94.8%,根系发达,茎上生长良好。本研究初步筛选出瓶内繁殖率高、生长良好、生根率高的优良无性系为A40,其根系发达、生长良好易移栽。
This paper reports on a study aimed at testing the effects of media with different hormone combinations on the clustered buds and rooting culture of Ochroma lagopus seedlings originating from seeds.This work then enabled the optimal medium formulation for each stage of culture to be identified.The results showed that 0.1%HgCl2 had the best disinfection effect over a duration of 15 minutes,and the pollution rate was only 6.6%.The explants grew best on S4 proliferation medium(MS+6-BA0.5 mg·L^(-1)+NAA0.2 mg·L^(-1))and had the best proliferation on this medium.The rooting rate on T2 rooting medium(1/2MS+IBA0.3 mg·L^(-1)+NAA0.1 mg·L^(-1))reached 94.8%,with well-developed roots and growth on stems.The best clone with high reproduction rate,good growth and high rooting rate was preliminarily selected as A40,which had developed good root systems,had good growth and was easily transplanted.
