摘要:为了建立夏栎(Quercus robur L.)器官发生及植株再生体系,以夏栎促萌枝条的幼嫩茎段为外植体,研究消毒方式、基本培养基的种类、外源激素的浓度及其配比等对其再生体系的影响。结果表明,夏栎幼嫩茎段以0.1%氯化汞溶液消毒6 min的成活率最高,为76.66%;基本培养基为WPM时生长状况最佳,在继代培养过程中,6-BA作用明显,以WPM培养基添加0.20 mg/L 6-BA对腋芽诱导分化效果最佳,以WPM培养基添加0.40 mg/L 6-BA对不定芽增殖效果最佳,增殖系数为4.2,接种在含0.25 mg/L IBA+0.10 mg/L NAA的生根培养基,生根率最高,为80.00%。本研究以夏栎一年生茎段为外植体,通过对其消毒、接种、茎段增殖、生根培养,可获得大量组培苗,为夏栎无性繁殖体系的建立、工厂化育苗及基因工程等应用提供理论基础和技术支持。
In order to establish the organogenesis and plant regeneration system of Quercus robur L.,the effects of different disinfection methods,different kinds of basic media,different concentrations of exogenous hormones and their ratio on its regeneration system were studied.The results showed that the highest percentage of success was 76.66%when the young stems of Quercus robur were disinfected with 0.1%mercuric chloride solution for 6 min.The growth status was the better when the basic medium was WPM.In the process of subculture,the effect of 6-BA was obvious.The effect of axillary bud differentiation was the best when 0.20 mg/L 6-BA was added to WPM medium.The effect of adventitious bud proliferation was the best when 0.40 mg/L 6-BA was added to WPM medium.The proliferation coefficient was 4.2.The rooting rate was the highest(80.00%)when the rooting medium was 0.25 mg/L IBA+0.10 mg/L NAA.In this study,the annual stem segments of Quercus robur were used as explants.Through disinfection,inoculation,stem proliferation and rooting culture,a large number of tissue culture seedlings could be obtained,which provided theoretical basis and technical support for the establishment of asexual propagation system,industrialized seedling raising and genetic engineering of Quercus robur.
