摘要:为了揭示核桃JAZ基因家族在核桃生长发育中的作用,本研究通过构建隐马尔科夫模型与转录组数据结合的方法,在核桃全基因组数据库搜索JAZ家族基因,对该家族进行理化性质、基因结构、保守基序、启动子元件、蛋白互作预测等生物信息学和低温胁迫下的转录表达分析。结果表明,核桃JAZ基因家族包含17个成员,蛋白长度范围为138~385个氨基酸,开放阅读框为417~1149 bp,等电点为8.47~10,全部为碱性不稳定亲水蛋白;含有典型的TIFY和jas 2个高度保守的结构域,聚类分析发现,该类基因可分为Group A、Group B、Group C和Group D 4个亚族。在低温胁迫48 h后经转录表达分析,JrJAZ1、JrJAZ3、JrJAZ4、JrJAZ5、JrJAZ7、JrJAZ9、JrJAZ15、JrJAZ16为差异表达基因,推测可能响应低温胁迫。本研究结果将为进一步研究该类基因的功能提供了重要线索和依据。
In order to reveal the role of the walnut JAZ gene family in the growth and development of walnuts,in this study,by constructing a Hidden Markov model combined with transcriptome data,the JAZ family genes were searched in the walnut genome database.Bioinformatics and transcriptional expression analysis was performed to study the gene structure,conserved motifs,promoter elements,and protein interaction prediction.We identified 17 JAZ members in walnut genome,protein length ranging from 138 to 385 amino acids,ORF length ranging from 417 to 1149,and the isoelectric points were from 8.47 to 10.all of them were alkaline unstable shydrophilic proteins.including two highly conserved domains,typically TIFY and jas,Cluster analysis found that the gene family can be divided into 4 subfamilies,Group A,Group B,Group C and Group D.Except to JrJAZ13 and JrJAZ16,there are abundant hormone signals and stress response regulatory elements.protein interaction predictions reveal that JrJAZs and other proteins.As well as within the same family of proteins,interactions can occur.Transcriptional expression analysis after 48 h of low temperature stress showed that,JrJAZ1,JrJAZ3,JrJAZ4,JrJAZ5,JrJAZ7,JrJAZ9,JrJAZ15 and JrJAZ16 were differentially expressed genes,which may respond to low temperature stress.The results of this study will provide important clues and basis for further studying the functions of these genes.
