摘要:【目的】重瓣棣棠花(Kerria japonica f.pleniflora)是我国常见的园林景观植物,其枝枯病时有发生,病、死枝的出现极大地降低了重瓣棣棠花的观赏性。准确鉴定重瓣棣棠花枝枯病的相关真菌,研究其形态学及分子系统学特征,是园林植物枝枯病防治理论的基础工作。【方法】采用单孢分离法和组织分离法获取重瓣棣棠花枝枯病相关真菌的菌株,并进行形态特征观察分析和联合多基因片段(ITS-act-rpb2-tef1-tub2)系统发育学研究。【结果】重瓣棣棠花病枝中分离出的18株真菌均鉴定为素贴壳囊孢(Cytospora diopuiensis),其分生孢子透明,腊肠形,无隔,光滑,壁薄,3.8~5.4μm×1.3~1.6μm,长宽比2.7~4。该菌亲缘关系较近的3种分别是东亚地区分布的掌状壳囊孢(C.palm),泰国壳囊孢(C.thailandica)和兴隆壳囊孢(C.xinglongensis)。【结论】研究揭示了重瓣棣棠花的枝枯病潜在病原菌为素贴壳囊孢,该菌首次在中国被发现,为中国新记录种。本研究首次描述了其无性型特征并讨论了亲缘关系较近的物种之间的关系,为病害防治和壳囊孢属的分类学研究提供了重要证据。
【Objective】Kerria japonica f.pleniflora is a common garden plant in China.However,the branch canker disease occurred recently,which greatly reduced ornamental values of the host.The aim of the present study is to identify the canker disease related Fungi based on morphology and modern molecular analyses.【Methods】Isolates were obtained from both conidia and diseased tissues.Five loci named ITS,act,rpb2,tef1 and tub2 were sequenced and combined to conduct the phylogenetical analyses.【Results】The 18 isolates was identified as Cytospora diopuiensis.Conidia of this fungus is hyaline,allantoid,smooth,aseptate,thin walled,3.8~5.4μm×1.3~1.6μm,with length width ratio from 2.7 to 4.Three phylogenetically close species of C.diopuiensis are C.palm,C.thailandica and C.xinglongensis,which are all distributed in east Asia.【Conclusion】The present study revealed the branch canker potential pathogen on K.japonica f.pleniflora as C.diopuiensis.This fungus is new to China and the asexual stage was firstly observed and illustrated.The accurate identification of the potential pathogen,and the phylogenetical relationship in the genus Cytospora provide the fundamental data to understand this disease and are helpful in disease management in the future.
