摘要:对农杆菌LBA4404转化柚木愈伤组织的适宜条件进行了优化.结果表明;LBA4404转化柚木愈伤组织的适宜预培养时间为11~15d,菌液光密度为0.5~0.8,共培养时间为13d,共培养的pH为5.6~6.2;重新悬浮的作用明显,乙酰丁香酮的作用不明显;不同无性系对LBA4404侵染的敏感性不一样,选择适宜的无性系可显著提高转化效率.
This paper deals with the genetic transformation of teak callus mediated by agro bacterium strain LBA4404. The results show that the optimized pre-culture time for callus inducement of stem section explants is 11 to 15 days ; that the OD600 value of bacterium inoculum solution is between 0. 5 and 0.8; and that the co-culture time is 13 days and pH value of co-culture medium is 5, 6 to 6.2. Re suspension of inoculum solution enhances the transformation efficiency but the addition of Actosyringone does not improve it, Different clones have different susceptibility to the infection of LBA4404 and clones with high susceptibility should be selected to do the transformation breeding of teak.
