摘要:以白银寿品种‘奇迹’花葶及花蕾为外植体,对其离体培养及快速繁殖技术进行了研究。结果表明,花蕾为适宜白银寿离体培养的外植体,适宜的愈伤组织诱导培养基为MS+6-BA 2.0 mg/L+KT 1.0 mg/L+NAA0.2 mg/L+蔗糖30 g/L+琼脂6.0 g/L,外植体脱分化速度快,愈伤组织质量好;最适分化培养基为:MS+6-BA1.0 mg/L+KT 0.5 mg/L+蔗糖30 g/L+琼脂6.0 g/L;适宜增殖培养基为MS+6-BA 0.4 mg/L+KT 0.4 mg/L+NAA0.1 mg/L+蔗糖30 g/L+琼脂6.0g/L,增殖系数达4.5;1/2MS+NAA 0.1 mg/L+蔗糖30 g/L+琼脂6.0 g/L是较适宜的生根培养基,生根率高且移栽成活率达到80%。利用花蕾为外植体,可以解决多肉植物因叶片含水量高而在灭菌后不易存活的问题,且采用花器官作为外植体不会损伤此类多肉植物的观赏部位,从而实现离体快速繁殖的目的。
Haworthia emelyae var. emelyae is a high valued succulent plant, it traditionally propagated via seed,leaf cuttings or crown division but the propagation rate is too low to satisfy commercial production. In order to improve the propagation rate, the tissue culture and rapid propagation systems were established for explant from scape and flower bud of Haworthia emelyae var. emelyae‘Miracle'. The results showed that the flower bud of Haworthia emelyae var. emelyae was the suitable explant in callus induction. The optimal callus formation rate was obtained on 0.65% agar-gelled Murashige and Skoog's(MS) basal medium supplemented with 2.0 mg/L6-benzylaminopurine(6-BA), 0.2 mg/L of a-naphthaleneacetic acid(NAA), and 1.0 mg/L kinetin(KT), meanwhile included 3% sucrose and 0.65% agar. The optimal shoot differentiation rate was generated on MS medium supplemented with 1.0 mg/L 6-BA and 0.5 mg/L KT. Our study showed that shoots proliferated well in medium containing 0.4 mg/L 6-BA, 0.4 mg/L KT and 0.1 mg/L NAA, meanwhile shoots rooted vigorously in half strength MS medium supplemented with 0.1 mg/L NAA. Finally regenerated plants were transferred to soil culture based on vermiculite and perlite(2:1), giving a survival rate of 80%. The culture method established for Haworthia emelyae var. emelyae‘Miracle'in this study can be applied for rapid propagation of Haworthia emelya.
